Studies exist on sperm cryopreservation of endangered Mohor gazelle (Gazella dama mhorr), but no work has been carried out yet on oocyte collection, fertilization and culture in this or related species. The purpose of this study was to develop a protocol for ovarian stimulation for the recovery of oocytes and subsequent IVMFC in the Mohor gazelle using frozen-thawed spermatozoa. Ovum pickup was performed after ovarian stimulation with a total dose of 5.28 mg of ovine FSH. A total of 35 oocytes were recovered from 56 punctured follicles (62%) (N = 6 females). Out of 29 cumulus-oocyte complexes matured in vitro, 3% were found at germinal vesicle stage, 7% at metaphase I, 21% were degenerated, and 69% advanced to metaphase II. Fertilization and cleavage rates of matured oocytes were 40 and 30%, respectively. Embryos cleaved in vitro up to the 6-8 cell stage but none progressed to the blastocyst stage, suggesting the existence of a developmental block and the need to improve culture conditions. Captive breeding provides the tool to rear sustained populations. Knowledge about biology of such species is critical for managing such breeding programmes. In this paper we look at the sex variation in calves surviving to 30 days in captive populations of two endangered polygynous gazelles, Gazella cuvieri and Gazella dama mhorr. We examined which of the following mother traits may affect sex allocation: her age, experience and inbreeding coefficient. Because twins are quite common in Cuvier's gazelle, we also analyze whether offspring sex is mediated by litter size in this gazelle. In both species the overall sex ratio did not differ from unity at the age of 30 days. These results support the suggestion that for a given population to be in equilibrium, a 1:1 calf sex ratio is to be expected, with females in good condition producing sons and females in poor condition producing daughters. We also found that offspring sex is not significantly related to the same mother traits in the two gazelles studied. Cuvier's gazelle findings showed that only maternal inbreeding influenced offspring sex, the less inbred the mother the higher the probability of producing sons. In contrast, none of the maternal traits studied were related to offspring sex in Mohor gazelle. Two non exclusive explanations are given to justify differences observed between these two species; one relate to a probable differential cost of producing either sex; the other refers to the actual homozygosis level in captive populations of each species. For Cuvier's gazelle litter size has no effect on offspring sex when the sex of the littermate is taken into account. This result fit predictions derived from Trivers-Willard model: mother in good conditions (less inbred) produce the sex with higher fitness returns (i.e., male), no matter if born at single or twins brood. Many variables are likely to affect progeny sex ratio. We have presently examined only a few. A severe inbreeding depression was encountered in a captive breeding program for Speke's gazelle. Unfortunately, the solution of simply avoiding inbreeding could not be implemented because the entire herd was descended from one import male and three import females. Because of this founder effect, it was impossible to avoid inbreeding. However, laboratory experiments with fruit flies and basic evolutionary theory indicate that animals can rapidly adapt to inbreeding by the selective elimination of the genes responsible for inbreeding depression. These experimental and theoretical results were translated into a breeding program for the Speke's gazelle. The first goal of the breeding program is a demographic goal: increase the total population size as rapidly as possible to the carrying capacity. The other goals all deal with genetic attributes of either parents or offspring: Both parents and offspring should be inbred, and both parents and offspring should have genes from as many different founding ancestors as possible. In this paper, we document that this breeding program does eliminate the inbreeding depression very rapidly, and moreover that the genetic goals of the program aid this elimination exactly as theory predicts. Furthermore, our analysis clearly shows that the gazelles suffered from an inbreeding depression rather than an outcrossing depression. The purpose of the present study was to evaluate various diluents for the cryopreservation of spermatozoa from three species of gazelles (genus Gazella) in a captive breeding program. The diluents compared were Tes (N-tris(hydroxymethyl)methyl-2 aminoethane sulfonic acid)-Tris with 5% egg yolk and 6% glycerol (TEST) and Triladyl, yolk-citrate, Tris-trehalose, and Tris-lactose-all of them with 20% egg yolk and 6% (Triladyl) or 8% glycerol. Semen was obtained by electroejaculation from 12 G. cuvieri, 12 G. dama, and 13 G. dorcas males. Samples with less than 50% motile sperm, positive endosmosis, or acrosome integrity were not used. Diluted samples were loaded into 0.25-ml straws, cooled slowly to 5؇C over 1.5 h (Ϫ0.16؇C/min), equilibrated at that temperature for 2 h, frozen in nitrogen vapors for 10 min, and plunged into liquid nitrogen. Subsamples were assessed fresh, after refrigeration-equilibration, after freezing and thawing, and 2 h after thawing. Differences were seen between diluents, with best overall recovery rates after freezing and thawing found with Triladyl, TEST, and Tris-trehalose in G. cuvieri, TEST in G. dama, and Triladyl and TEST in G. dorcas. Differences were observed between species in the ability to withstand freezing and thawing, with best results seen in G. dorcas, intermediate results in G. dama, and worst results in G. cuvieri. These differences were inversely related to the average values of inbreeding of these populations. The effective number of founders, ancestors and founder genomes was found to be 3.42, 3, and 1.44 for the studied reference population.
