Bionomics data are consisted of breeding places, resting places and feeding habits are from direct observation. Interviews and questionnaires were conducted to gain information about respondent behavior. Descriptive and qualitative data were then analyzed. The breeding places of Anopheles sp. were mostly found in swampy areas. Based on the feeding habit, the feeding activity of Anopheles sp. inside the house has only one biting peak at 23.00-02.00 WIT, while outside the house, biting peaks occurred at 21.00-22.00 WIT and 00.00-01.00 WIT. Resting place data shown that Anopheles sp. mostly found in piles of clothes and shoe racks. This study aimed to identify the breeding place distribution and bionomics of Anopheles spp. in Runut village, Sikka district, East Nusa Tenggara. Methods: A descriptive observational study was conducted in Runut village, Waigete sub- district, in April 2018. Anopheles spp. larvae were collected in 7 suspected breeding places consisting of 2 rice fields, 3 fish ponds, and 2 puddles. Species-specific behaviours, such as host feeding preferences, impact the dynamics of malaria transmission and the effectiveness of vector control interventions. To examine species-specific host attraction and feeding behaviours, a Latin square design was used to compare Anopheles mosquitoes attracted to human, cow, and goat-baited tents. Anopheles mosquitoes were collected hourly from the inside walls of each baited tent. Species were morphologically and then molecularly identified using rDNA ITS2 sequences. The head and thorax of individual specimens were analysed for Plasmodium DNA using PCR. Bloodmeals were identified using a multiplex PCR. A total of 1024, 137, and 74 Anopheles were collected over 12 nights in cow, goat, and human-baited tents, respectively. This study was an observational analytic research by using adult mosquito specimens. The mosquitoes were caught using indoor net trapping and outdoor cow baited net trapping started at 18.45 to 24.00 for six catching times. Outdoor cow baited net trapping caught 61 mosquitoes from seven different species namely An. aconitus, An. annularis, An. barbirostris, An. indefinitus, An. kochi, and An. vagus. Human bait trap only trapped one species, An. indefinites.
