the major risk factors in Non-Communicable Disease. Free radicals from cigarette smoke can cause oxidative stress. The oxidative insults can lead to red blood cell (RBC) senescence and are involved in the clearance of red blood cells. The objective of the present study is to assess the association between smoke, oxidative stress evaluated with serum Malondialdehyde (MDA) level and phosphatidylserine (PS) externalization (biomarker of RBC senescence) evaluated with annexin V binding. A total of sixty-four male volunteers aged 25-60 years old were recruited in this study. MDA was measured by colorimetric method. Annexin V binding was detected by flow cytometry. Our results show that there was a significant increase in MDA levels in cigarette smokers as compared to non-smokers (p < 0.001). However, there was no significant different between annexin V binding (% gate) in cigarette smokers and non-smokers (p = 0.978). These results provide evidence of free radical from smoking is associated with oxidative damage to erythrocytes. However, our results suggest that PS externalization is unlikely to have a role in RBC senescence pathway of stressed erythrocytes from cigarette smoke. The other biomarker of RBC senescence should be determined on cigarette smoker erythrocytes. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=malondialdehyde" title="malondialdehyde">malondialdehyde</a>, <a href="https://publications.waset.org/abstracts/search?q=phosphatidylserine" title=" phosphatidylserine"> phosphatidylserine</a>, <a href="https://publications.waset.org/abstracts/search?q=RBC%20senescence" title=" RBC senescence"> RBC senescence</a>, <a href="https://publications.waset.org/abstracts/search?q=annexin%20V" title=" annexin V"> annexin V</a> </p> <a href="https://publications.waset.org/abstracts/28190/effect-of-oxidative-stress-from-smoking-on-erythrocyte-phosphatidylserine-externalization" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/28190.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">437</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">4</span> Rauvolfine B Isolated from the Bark of Rauvolfia reflexa (Apocynaceae) Induces Apoptosis through Activation of Caspase-9 Coupled with S Phase Cell Cycle Arrest</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Mehran%20Fadaeinasab">Mehran Fadaeinasab</a>, <a href="https://publications.waset.org/abstracts/search?q=Hamed%20Karimian"> Hamed Karimian</a>, <a href="https://publications.waset.org/abstracts/search?q=Najihah%20Mohd%20Hashim"> Najihah Mohd Hashim</a>, <a href="https://publications.waset.org/abstracts/search?q=Hapipah%20Mohd%20Ali"> Hapipah Mohd Ali </a> </p> <p class="card-text"><strong>Abstract:</strong></p> In this study, three indole alkaloids namely; rauvolfine B, macusine B, and isoreserpiline have been isolated from the dichloromethane crude extract of Rauvolfia reflexa bark (Apocynaceae). The structural elucidation of the isolated compounds has been performed using spectral methods such as UV, IR, MS, 1D, and 2D NMR. Rauvolfine B showed anti proliferation activity on HCT-116 cancer cell line, its cytotoxicity induction was observed using MTT assay in eight different cell lines. Annexin-V is serving as a marker for apoptotic cells and the Annexin-V-FITC assay was carried out to observe the detection of cell-surface Phosphatidylserine (PS). Apoptosis was confirmed by using caspase-8 and -9 assays. Cell cycle arrest was also investigated using flowcytometric analysis. rauvolfine B had exhibited significantly higher cytotoxicity against HCT-116 cell line. The treatment significantly arrested HCT-116 cells in the S phase. Together, the results presented in this study demonstrated that rauvolfine B inhibited the proliferation of HCT-116 cells and programmed cell death followed by cell cycle arrest. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=apocynacea" title="apocynacea">apocynacea</a>, <a href="https://publications.waset.org/abstracts/search?q=indole%20alkaloid" title=" indole alkaloid"> indole alkaloid</a>, <a href="https://publications.waset.org/abstracts/search?q=apoptosis" title=" apoptosis"> apoptosis</a>, <a href="https://publications.waset.org/abstracts/search?q=cell%20cycle%20arrest" title=" cell cycle arrest"> cell cycle arrest</a> </p> <a href="https://publications.waset.org/abstracts/13403/rauvolfine-b-isolated-from-the-bark-of-rauvolfia-reflexa-apocynaceae-induces-apoptosis-through-activation-of-caspase-9-coupled-with-s-phase-cell-cycle-arrest" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/13403.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">334</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">3</span> Inflammatory Alleviation on Microglia Cells by an Apoptotic Mimicry</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Yi-Feng%20Kao">Yi-Feng Kao</a>, <a href="https://publications.waset.org/abstracts/search?q=Huey-Jine%20Chai"> Huey-Jine Chai</a>, <a href="https://publications.waset.org/abstracts/search?q=Chin-I%20Chang"> Chin-I Chang</a>, <a href="https://publications.waset.org/abstracts/search?q=Yi-Chen%20Chen"> Yi-Chen Chen</a>, <a href="https://publications.waset.org/abstracts/search?q=June-Ru%20Chen"> June-Ru Chen</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Microglia is a macrophage that resides in brain, and overactive microglia may result in brain neuron damage or inflammation. In this study, the phospholipids was extracted from squid skin and manufactured into a liposome (SQ liposome) to mimic apoptotic body. We then evaluated anti-inflammatory effects of SQ liposome on mouse microglial cell line (BV-2) by lipopolysaccharide (LPS) induction. First, the major phospholipid constituents in the squid skin extract were including 46.2% of phosphatidylcholine, 18.4% of phosphatidylethanolamine, 7.7% of phosphatidylserine, 3.5% of phosphatidylinositol, 4.9% of Lysophosphatidylcholine and 19.3% of other phospholipids by HPLC-UV analysis. The contents of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in the squid skin extract were 11.8 and 28.7%, respectively. The microscopic images showed that microglia cells can engulf apoptotic cells or SQ-liposome. In cell based studies, there was no cytotoxicity to BV-2 as the concentration of SQ-liposome was less than 2.5 mg/mL. The LPS induced pro-inflammatory cytokines, including tumor necrosis factor-alpha (TNF-伪) and interleukin-6 (IL-6), were significant suppressed (P < 0.05) by pretreated 0.03~2.5mg/ml SQ liposome. Oppositely, the anti-inflammatory cytokines transforming growth factor-beta (TGF-尾) and interleukin-10 (IL-10) secretion were enhanced (P < 0.05). The results suggested that SQ-liposome possess anti-inflammatory properties on BV-2 and may be a good strategy for against neuro-inflammatory disease. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=apoptotic%20mimicry" title="apoptotic mimicry">apoptotic mimicry</a>, <a href="https://publications.waset.org/abstracts/search?q=neuroinflammation" title=" neuroinflammation"> neuroinflammation</a>, <a href="https://publications.waset.org/abstracts/search?q=microglia" title=" microglia"> microglia</a>, <a href="https://publications.waset.org/abstracts/search?q=squid%20processing%20by-products" title=" squid processing by-products"> squid processing by-products</a> </p> <a href="https://publications.waset.org/abstracts/78159/inflammatory-alleviation-on-microglia-cells-by-an-apoptotic-mimicry" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/78159.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">483</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2</span> Natural Honey and Effect on the Activity of the Cells</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Abujnah%20Dukali">Abujnah Dukali</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Natural honey was assessed in cell culture system for its anticancer activity. Human leukemic cell line HL 60 was treated with honey and cultured for 5 days and cytotoxicity was calculated by MTT assay. Honey showed cytotoxicity with CC50 value of 174.20 碌g/ml. Radical modulation activities was assessed by lipid peroxidation assay using egg lecithin. Honey showed antioxidant activity with EC50 value of 159.73 碌g/ml. In addition, treatment with HL60 cells also resulted in nuclear DNA fragmentation, as seen in agarose gel electrophoresis. This is a hallmark of cells undergoing apoptosis. Confirmation of apoptosis was performed by staining the cells with Annexin V and FACS analysis. Apoptosis is an active, genetically regulated disassembly of the cell form within. Disassembly creates changes in the phospholipid content of the cytoplasmic membrane outer leaflet. Phosphatidylserine (PS) is translocated from the inner to the outer surface of the cell for phagocytic cell recognition. The human anticoagulant, annexin V, is a Ca2+-dependent phospholipid protein with a high affinity for PS. Annexin V labeled with fluorescein can identify apoptotic cells in the population It is a confirmatory test for apoptosis. Annexin V-positive cells were defined as apoptotic cells. Since honey shows both antioxidant activity and cytotoxicity at almost the same concentration, it can prevent the free radical induced cancer as prophylactic agent and kill the cancer cells by apoptotic process as a chemotherapeutic agent. Everyday intake of honey can prevent the cancer induction. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=anticancer" title="anticancer">anticancer</a>, <a href="https://publications.waset.org/abstracts/search?q=cells" title=" cells"> cells</a>, <a href="https://publications.waset.org/abstracts/search?q=DNA" title=" DNA"> DNA</a>, <a href="https://publications.waset.org/abstracts/search?q=honey" title=" honey"> honey</a> </p> <a href="https://publications.waset.org/abstracts/44196/natural-honey-and-effect-on-the-activity-of-the-cells" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/44196.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">206</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">1</span> Satureja bachtiarica Bunge Induce Apoptosis via Mitochondrial Intrinsic Pathway and G1 Cell Cycle Arrest</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Hamed%20Karimian">Hamed Karimian</a>, <a href="https://publications.waset.org/abstracts/search?q=Noraziah%20Nordin"> Noraziah Nordin</a>, <a href="https://publications.waset.org/abstracts/search?q=Mohamad%20Ibrahim%20Noordin"> Mohamad Ibrahim Noordin</a>, <a href="https://publications.waset.org/abstracts/search?q=Syam%20Mohan"> Syam Mohan</a>, <a href="https://publications.waset.org/abstracts/search?q=Mahboubeh%20Razavi"> Mahboubeh Razavi</a>, <a href="https://publications.waset.org/abstracts/search?q=Najihah%20Mohd%20Hashim"> Najihah Mohd Hashim</a>, <a href="https://publications.waset.org/abstracts/search?q=Happipah%20Mohd%20Ali"> Happipah Mohd Ali</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Satureja bachtiarica Bunge is a perennial medicinal plant belonging to the Lamiaceae family and endemic species in Iran. Satureja bachtiarica Bunge with the local name of Marzeh koohi is edible vegetable use as flavoring agent, anti-bacterial and to relieve cough and indigestion. In this study, the anti-cancer effect of Satureja bachtiarica Bunge on the MDA-MB-231 cell line as an Breast cancer cell model has been analyzed for the first time. Satureja bachtiarica Bunge was extracted using different solvents in the order of increasing polarity. Cytotoxicity activity of hexane extract of Satureja bachtiarica Bunge (SBHE) was observed using MTT assay. Acridine orange/Propidium iodide staining was used to detect early apoptosis; Annexin-V-FITC assay was carried out to observe the detection of cell-surface Phosphatidylserine (PS), with Annexin-Vserving as a marker for apoptotic cells. Caspase 3/7, 8 and-9 assays showed significantly activation of caspase-9 where lead intrinsic mitochondrial pathway. Bcl-2/Bax expressions and cell cycle arrest were also investigated. SBHE had exhibited significantly higher cytotoxicity against MDA-MB-231 Cell line compare to other cell lines. A significant increase in chromatin condensation in the cell nucleus was observed by fluorescence analysis. Treatment of MDA-MB-231 cells with SBHE encouraged apoptosis, by down-regulating Bcl-2 and up-regulating Bax, which lead the activation of caspase 9. Moreover, SBHE treatment significantly arrested MDA-MB-231 cells in the G1 phase. Together, the results presented in this study demonstrated that SBHE inhibited the proliferation of MDA-MB-231 cells, leading cell cycle arrest and programmed cell death, which was confirmed to be
