In Expt 1 six P\l=e`\reDavid's deer and 12 red deer hinds were treated during the breeding season with an intravaginal progesterone-impregnated controlled internal drug release device (CIDR) for 14 days, with 200 iu pregnant mares' serum gonadotrophin (PMSG) administered 72 h before the device was withdrawn and eight injections of ovine FSH given at 12 h intervals starting at the time of PMSG administration. Oestrous behaviour began one day after CIDR device withdrawal (P\l=e`\re David's deer: 24.00 \ m=+-\ 2.32 h; red deer: 24.60 \ m=+-\ 2.23 h). The duration of oestrus was greater in P\l=e`\re David's deer than in red deer (17.50 \ m=+-\ 1.43 h and 8.25 \ m=+-\ 3.25 h, respectively, P<0.001). The peak LH surge of P\l=e`\reDavid's deer was 68.65 \ m=+-\ 4.74 ng ml\m=-\1 occurring 29.00 \ m=+-\ 2.41 h after removal of the CIDR devices. In comparison, the peak LH surge in red deer was 17.09 \ m=+-\ 3.64 ng ml \ m=-\ 1 (P<0.001), occurring 24.00 \ m=+-\ 0.00 h after CIDR device withdrawal. Pre-surge concentrations of LH were also greater (P<0.001) in P\l =e`\re David's deer (1.37 \ m=+-\ 0.11 ng ml\m=-\1) than in red deer hinds (0.41 \ m=+-\ 0.02 ng ml \m=-\1). Oestradiol concentrations increased steadily during FSH treatment in both species and reached a maximum of 15.60 \ m=+-\ 2.22 pg ml\m=-\1at 21.00 \ m=+-\ 6.20 h after CIDR device withdrawal in P\l=e`\reDavid's deer and 26.14 \ m=+-\ 6.20 pg ml \ m=-\ 1 at 20.40 \ m=+-\7.30 h before CIDR device removal in red deer. All hinds were inseminated in utero 36 h after CIDR device withdrawal with 25 \ m=x\ 106 frozen\p=n-\thawedP\l =e`\re David's deer spermatozoa. Embryos were recovered by laparotomy 5, 6 and 7 days after insemination. A total of six embryos (three P\l=e`\reDavid's deer \ m=x\red deer and three P\l=e`\reDavid's deer \ m=x\P\l=e`\reDavid's deer) were collected. One pregnancy was confirmed following the transfer of all embryos to synchronized red deer recipients. This pregnancy was terminated naturally between days 70 and 78. This is the first report of an interspecific transfer of hybrid embryos in a cervid and of the comparative superovulation response in two related species. The results indicate that treatment with PMSG and ovine FSH can be used in embryo transfer programmes for these species. Unfortunately, reproductive success has been poor despite breeding recommendations for almost every reproductively viable bear by the Species Survival Plant. Assisted reproductive technologies could complement breeding efforts by overcoming the challenges of behavioral incompatibilities and deficiencies, facilitating genetic management and increasing cub production. The goal of this study was to artificially inseminate a female polar bear after inducing ovarian activity and ovulation with exogenous hormones (equine chorionic gonadotropin and porcine luteinizing hormone). Fresh semen collected from an adult male via electroejaculation/urethral catheterization was used for the insemination. Fecal steroid monitoring indicated that the female ovulated following the exogenous hormone treatment. Progestin concentrations increased in late summer, at the time implantation was expected to occur; however, no cubs were produced. Pregnancy rates using these two transvaginal AI methods were also compared in a commercial herd of fallow does. The estrous cycles of fallow n 53, red n 27 and white-tailed n 27 deer were synchronized using half of a Norgestomet ear implant inserted for 14 days. Following implant removal, does were arti®cially ®x-time inseminated with frozen-thawed semen using either a 0.5 ml standard French-style speculum-guided AI gun or the Gourley Scope method of AI. The time-frame (i.e., duration in minutes) for the AI procedures performed on each animal and the location of semen deposition was recorded. To assess effectiveness of AI, transrectal ultrasonography was conducted in fallow deer 45 days post-AI for pregnancy diagnosis. In fallow does and red deer hinds, use of the Gourley Scope (96.3 and 100%, respectively) enabled a greater P < 0:05 percentage of intrauterine inseminations compared to the speculum-guided AI gun (73.1 and 42.9%, respectively). In contrast for white-tailed does, both the speculum-guided AI gun and the Gourley Scope resulted in a similar proportion of intrauterine inseminations (92.3 and 100%, respectively). The duration for performing the two transvaginal AI techniques were also similar in red and white-tailed deer. However in fallow does, AI with the aid of the Gourley Scope took longer P < 0:05 to conduct than the speculum-guided AI gun (6:6 AE 0:72 and 4:3 AE 0:34 min, respectively). The pregnancy rate using both methods of AI combined was overall 32.1% for fallow does. The total percentage of fallow does that conceived with the speculum-guided AI gun was 42.3% whereas the pregnancy rate using the Gourley method of AI was only 22.2% P < 0:05.
