This study was aimed at investigating the antisickling activity of the hydro-ethanolic extract of a combination of the leaves of J. secunda, J. gossypiifolia and P. nigrescens. These three plants species were used in the Ivorian traditional herbal medicine. Preliminary phytochemistry was carried out using standard methods. As for the sickling reversal test, the Hb SS blood sickling was induced with 2% sodium metabisulfite. After 120 minutes of incubation, the plant extract was added. Every 30 minutes for a period of 120 minutes, a drop of the prepared solution was observed at (40×) magnification and the percentage of reversion calculated. The phytochemical analysis revealed the presence of Alkaloids, Flavonoids, Polyphenols, Catechic Tannins, Sterols and Polyterpenes. The results of the reversal test showed that the percentage of sickling reversal effect of the combination of the plants (75.00 ± 4.33 b,c ) was highly superior than the negative control (10.17 ± 0.55 d ) but was significantly (p < 0.05) the same as the percentage of the individual plant (J. secunda: 83.50 ± 2.33 a ; J. gossypiifolia: 78.00 ± 3.67 b,c and P. nigrescens: 77.83 ± 2.89 b,c ) and the positive control (80.66 ± 2.22 a,b ). From the results, the extracts J. secunda, J. gossypiifolia and P. nigrescens have shown to be therapeutically beneficial to the population. AS and SS red blood cells are subjected to hyposmotic impact with decreasing concentrations of NaCl solution. All the aqueous extracts of the six plants showed a better contribution in erythrocyte osmotic resistance from the concentration of 5 mg/mL to 1 mg/mL, except the extract at 5 mg/mL of Raphiostylis beninensis that caused hemolysis of both red blood cells AS and SS. The extract at 1 mg/mL of Raphiostylis beninensis and the extracts at 5mg/mL of Xylopia aethiopica and Croton zambesicus showed a high hemolysis inhibition of red blood cells AS and SS. The extracts of Lonchocarpus cyanescens showed moderate hemolysis inhibition of SS red blood cells while extracts of Uvaria chamae inhibited highly the hemolysis of AS red blood cells. As for the roots of Morinda lucida, only the extract at 5mg/mL highly inhibited the hemolysis of the AS red blood cells. This protocol seems appropriate to work with both AS and SS blood because the comparative effects of each tested extract on hemolysis of AS and SS blood showed a good correlation coefficient of Pearson (1 or-1). The study was conducted to document some of the medicinal plants used in the management of SCD with the aim of identifying possible drug leads from the phytomedicine of the area. A semi-structured questionnaire was used to obtain information on the use of herbal remedies for the management of SCD. A total of 32 respondents were interviewed in this study. twenty of them were males while twelve were females with the age range of 25 to 70 years. A total of 44 plant species belonging to 44 genera and 30 families were identified to be useful for the management of SCD. The most prominent among these plant families (genera) are Anacardiaceae ( Anacardium occidentalis, Mangifera indica, Spondias mombin ), Euphorbiaceae (C roton lobatus, Jatropha gossypifolia, Plukeneta conophora ) and Fabaceae ( Cajanus cajan, Pterocarpus osun, Cassia siamea ) with three species each. Eighty percent (80%) of the 32 respondents interviewed said that their clients did not report any side effect and that the herbs were either collected from the forest (where available) or purchased from the herb markets. In all, the commonest plant species among the recipes given by the respondents according to their Use Mention Index (UMI) include: Elaeis guineense Jacq., Arecaceae (0.124); Khaya grandifoliola C.DC., Meliaceae (0.094); Carica papaya L., Caricaceae(0.094); Hymenocardia acida Tul., Phyllanthaceae (0.094); Mangifera indica L., Anacardiaceae (0.094); Parquetina nigrescens (Afzel.) Rich., Annonaceae (0.094).The main method of preparation is by decoction and in most cases, the preparation is taken orally twice to thrice daily. All the plants identified in this work have been used by most of the herbalists and adjudged to be efficacious in SCD management. Materials and Methods: Information was obtained using semi-structured questionnaires and open interview from respondents consisting of herb sellers, traditional medical practitioners, artisans and traders in two local government areas of Ife. Two recipes from the survey were selected for antisickling studies. Aqueous extract of each recipe was obtained by boiling the constituents in water for 1 h while the hydro ethanolic extracts were obtained by maceration in 70% ethanol for 72 h. Inhibitory and reversal antisickling properties were assessed using sodium metabisulphite as deoxygenating agent, vanillic acid (inhibitory agent), para-hydroxybenzoic (reversal agent) acid as positive controls while phosphate buffered saline was employed as negative control. The lower strata of the population living in developing countries rely heavily on traditional medicine due to their cultural alignment as well as their inability to afford the cost of treatment offered by orthodox medical practitioners. Myths fuelled by cultural beliefs are also issues to contend with. For example, childhood deaths within a family were always attributed to some evil forces in the traditional African setting.
