reactivity of tocopherols with 2,2-diphenyl-1-picrylhydrazyl (DPPH) was studied in model systems in order to establish a method for quantifying vitamin E in plant oils. The method was optimized with respect to solvent composition of the assay medium, which has a large influence on the course of reaction of tocopherols with DPPH. The rate of reaction of α-tocopherol with DPPH is higher than that of γ-tocopherol in both protic and aprotic solvents. In ethyl acetate, routinely applied for the analysis of antioxidant potential (AOP) of plant oils, reactions of tocopherols with DPPH are slower and concentration of tocopherols in the assay has a large influence on their molar reactivity. In 2-propanol, however, two electrons are exchanged for both α-and γ-tocopherols, independent of their concentration. 2-propanol is not toxic and is fully compatible with polypropylene labware. The chromatographically determined content of tocopherols and their molar reactivity in the DPPH assay reveal that only tocopherols contribute to the AOP of sunflower oil, whereas the contribution of tocopherols to the AOP of linseed oil is 75%. The DPPH assay in 2-propanol can be applied for rapid and cheap estimation of vitamin E content in plant oils where tocopherols are major Herein, rapid, sensitive and precise method for the determination of all type of tocopherols in different vegetable oils includinhg sunflower oil, olive oil and extra virgin olive oil is reported. Reverse Phase High Performance Liquid Chromatography (RP-HPLC) with UV detector was used for tocopherol determination. A simple, quick and sensitive method to estimate the antioxidant quantity is proposed. In this method oils were diluted in methanol and injected directly into column (no saponification procedure). Methanol and water (98:2) mixture was used as mobile phase. Three tocopherols (α, γ and δ) were detected at 292 nm wavelength with UV detector. Method had good limit of detection (LOD) (7ng/g) and reproducibility (C.V% 0.9, 0.8 and 0.4 for α, γ and δ tocopherols, respectively). Result showed the best source for α-tocopherol was sun flower oil (146.65±1.7mg/kg). Oil richest in δ-tocopherol was olive oil and δ-tocopherol was absent in extra virgin olive oil. The current study suggest the new parameter (ratio of α/γ) as first screening indicator of authenticity of purity of extra virgin olive oil for differentiation of various cultivators of same generic developed a method for the determination of tocopherols in different edible oils by RP-HPLC, using a FLD detector, a Poroshell column as a stationary phase, and a mixture of acetonitrile, methanol and water (5:4.5:0.5) as mobile phase (flow rate 0.75 ml/min). The structure of the compounds was identical, so it could not be separated by reverse phase liquid chromatography. The obtained peaks show an important overlapping and we have used chemometric tools to get a calibration model, which assumes a linear behavior, i.e., the total signal is the sum of individual signals of components. Recovery percentages were between 90.3% and 95.7%. The method has been successfully applied to edible oils and dietary supplements. The chromatographic separation occurred isocratically with methanol-water [96:4%v/v] at 0.9 ml/min flow rate. Tocopheryl acetate was the internal standard and alpha-tocopherol was eluted at 7.87 min. Free fatty acids value [FFAVs], peroxide value [PV], iodine value [IV] and saponification values [SV] were determined as quality parameters. Calibration curve was linear [r2 0.9969] and the method was precise with relative standard deviation of 0.35% and mean recovery, 87.39%. Alpha-tocopherol concentration ranged from 0-9.22 mg/100g with the highest in Tropical sunflower oil [9.22 mg/100g] and the lowest [1.16 mg/100g] in Laziz oil.
