The TBS STDP-like protocol produced in the adult significant changes in the synaptic gain of the electrically activated thalamic pathway, but the statistical significance of the functional correlates was detectable mostly at the population level. Comparison of our observations with the literature leads us to re-examine the experimental status of spike timing-dependent potentiation in adult cortex. We have observed in cats that these shifts occur in a pattern that is highly convergent, even among receptive fields that are separated by large distances before inactivation. Here we show, using a computational model of primary visual cortex, that the observed convergent shifts are inconsistent with the common assumption that the underlying intracortical connection plasticity is dependent on the temporal correlation of pre-and postsynaptic action potentials. The shifts are, however, consistent with the hypothesis that this plasticity is dependent on the temporal order of pre-and postsynaptic action potentials. Therefore, it is important to take into account the variety of striatal neurons to fully understand the ability of striatum to develop long-term plasticity. While research over the past 2 decades has addressed these questions, several studies within the past 5 years have shown that synapses undergo long-term depression (LTD) or longterm potentiation (LTP) depending on the order of activity in the pre-and postsynaptic cells. This process has been referred to as spike-timing dependent plasticity (STDP). Here we discuss the experimental data on STDP, and develop models of the mechanisms that may underlie it. Specifically, we examine whether the standard model of LTP and LTD in which high and low levels of Ca 2þ produce LTP and LTD, respectively, can also account for STDP. We conclude that the standard model can account for a type of STDP in which, counterintuitively, LTD will be observed at some intervals in which the presynaptic cell fires before the postsynaptic cell. This form of STDP will also be sensitive to parameters such as the presence of an afterdepolarization following an action potential. Indeed, the sensitivity of this type of STDP to experimental parameters suggests that it may not play an important physiological role in vivo. The present study investigated STDP in vivo using alternative procedures for modulating dopaminergic inputs. Postsynaptic potentials (PSP) were evoked in intracellularly recorded spiny neurons by electrical stimulation of the contralateral motor cortex. PSPs often consisted of up to three distinct components, likely representing distinct cortico-striatal pathways. After baseline recording, bicuculline (BIC) was ejected into the superior colliculus (SC) to disinhibit visual pathways to the dopamine cells and striatum. Repetitive cortical stimulation (~60; 0.2Hz) was then paired with postsynaptic spike discharge induced by an intracellular current pulse, with each pairing followed 250ms later by a light flash to the contralateral eye (n=13). Changes in PSPs, measured as the maximal slope normalized to 5-min pre, ranged from potentiation (~120%) to depression (~80%). The determining factor was the relative timing between PSP components and spike: PSP components coinciding or closely following the spike tended towards potentiation, whereas PSP components preceding the spike were depressed. Importantly, STDP was only seen in experiments with successful BIC-mediated disinhibition (n=10). Cortico-striatal high-frequency stimulation (50 pulses at 100Hz) followed 100ms later by a light flash did not induce more robust synaptic plasticity (n=9). However, an elevated post-light spike rate correlated with depression across plasticity protocols (R2=0.55, p=0.009, n=11 active neurons). In the most basic formulation, STDP provides a fundamental unit-a spike pair-for quantifying the induction of long-term changes in synaptic strength. However, many factors, both pre-and postsynaptic, can affect synaptic transmission and integration, especially when multiple spikes are considered. Here we review the experimental evidence for multiple types of nonlinear temporal interactions in STDP, focusing on the contributions of individual spike pairs, overall spike rate, and precise spike timing for modification of cortical and hippocampal excitatory synapses. We discuss the underlying processes that determine the specific learning rules at different synapses, such as postsynaptic excitability and short-term depression. Several models of STDP have been proposed, and differ by their implementation, and in particular how synaptic weights saturate to their minimal and maximal values. We analyze here kinetic models of transmitter-receptor interaction and derive a series of STDP models. In general, such kinetic models predict progressive saturation of the weights. Various forms can be obtained depending on the hypotheses made in the kinetic model, and these include a simple linear dependence on the value of the weight ("soft bounds"), mixed soft and abrupt saturation ("hard bound"), or more complex forms. We analyze in more detail simple soft-bound models of Hebbian and anti-Hebbian STDPs, in which nonlinear spike interactions (triplets) are taken into account. We show that Hebbian STDPs can be used to selectively potentiate synapses that are correlated in time, while anti-Hebbian STDPs depress correlated synapses, despite the presence of nonlinear spike interactions. This correlation detection enables neurons to develop a selectivity to correlated inputs. We also examine different versions of kinetics-based STDP models and compare their sensitivity to correlations.
