margin: 0px 0px 26px; text-align: justify; word-break: break-all;"> 论文第一作者、中山大学附属第一医院关节外科助理研究员郑霖力表示，类风湿关节炎是一种以滑膜炎症为病理基础的自身免疫性疾病，最终可能导致关节畸形。类风湿关节炎患者的滑膜炎症表现出多样性，主要体现在免疫细胞的浸润和基质细胞的活化。然而，目前尚不清楚参与该疾病的关键细胞亚群。</p> <p style=";; margin: 0px 0px 26px; text-align: justify; word-break: break-all;"> 此外，滑膜中的基质细胞与免疫细胞共同参与类风湿关节炎的滑膜炎症反应。类风湿关节炎滑膜中存在一个高度复杂的免疫调控网络，深入研究滑膜微环境中免疫细胞和基质细胞的组成、分子特征、空间分布及其相互作用，对于揭示类风湿关节炎的发病机制具有重要意义。</p> <p style=";; margin: 0px 0px 26px; text-align: justify; word-break: break-all;"> 该研究对类风湿关节炎患者和骨关节炎患者的滑膜组织进行了单细胞转录组、T细胞受体及空间转录组测序，识别出包括免疫细胞和固有细胞在内的45个细胞亚群，详细绘制了类风湿关节炎患者滑膜的单细胞和空间转录组图谱。研究发现，ITGA5<sup>+</sup>滑膜成纤维细胞是类风湿关节炎患者滑膜中参与疾病进程的关键细胞，这类细胞在基质重塑中发挥作用，且高表达POSTN、COL3A1、CCL5和TGFB1等基因，富集于免疫调控相关通路。</p> <p style=";; margin: 0px 0px 26px; text-align: justify; word-break: break-all;"> ITGA5<sup>+</sup>滑膜成纤维细胞在类风湿关节炎活动期患者及淋巴细胞浸润型滑膜炎中上调，可能与类风湿关节炎的治疗抵抗性相关。这一细胞亚群可能源自DPP4<sup>+</sup>PI16<sup>+</sup>CD34<sup>+</sup>成纤维细胞的分化。ITGA5<sup>+</sup>滑膜成纤维细胞可能在滑膜炎症的早期阶段出现，并通过分泌TGF-β1诱导幼稚CD4<sup>+</sup> T细胞向CXCL13<sup>hi</sup> PD-1<sup>hi</sup> TPHs方向分化。关节腔内注射ITGA5<sup>+</sup>滑膜成纤维细胞会加重胶原诱导关节炎小鼠的滑膜炎症和骨破坏。</p> <p style=";;
