color: #333333; font-family: 宋体; line-height: 20px;"> <b></b></td> </tr> </table> <p>  </p> <p style="text-indent:2em"> 2024年10月17日，浙江大学药学院南科望研究员、建筑工程学院付浩然研究员、化学工程与生物工程学院关怡新教授团队合作在<em>Matter</em>期刊上发表一篇题为“Drug Origami: A Computation-Guided Approach for Customizable Drug Release Kinetics of Oral Formulations”的研究成果。</p> <p style="text-indent:2em"> 该研究创新性地提出了“折纸药物（<em>Drug origami</em>）”，通过精确设计包含屏障层和药物释放层的双层薄膜的折叠方式，实现药物的分阶段可控释放。该方法展示了未来个性化药物制剂的可能性，尤其是能够根据患者的具体病情节律进行个性化的药物释放调控。</p> <p style="text-indent:2em"> 浙江大学化工学院博士生黄浩和建工学院博士生张浩宇为本文的第一作者。</p> <p style="text-indent:2em"> 患者依从性问题长期困扰医学界，尤其是在慢性病治疗中，患者往往因需要频繁用药而无法完全遵从医生的用药建议，导致治疗效果大打折扣。尽管现有的许多缓释口服药物制剂具有延长治疗效果的优势，但它们在顺应生物节律和特定病理特征方面仍存在显著的局限性，同时在药物释放曲线的调控上缺乏灵活性，难以满足个体化的治疗需求。</p> <p style="text-indent:2em"> 为了解决上述问题，研究人员创新性提出“折纸药物（<em>Drug origami</em>）”，其核心理念在于利用可折叠的双层薄膜，其中包含一个屏障层和一个药物释放层，总厚度小于500微米；通过设计屏障层厚度和药物折叠方式，实现药物的分阶段可控释放。在此基础上，团队建立了计算模型精确预测折纸药物的释放动力学。</p> <p style="text-indent:2em"> 此外，研究还建立了折纸药物的反向设计框架，可根据特定药物释放曲线，设计相应的药物折叠方式（如图1）。体外和体内实验结果显示，计算模型和数值模拟的预测与实验数据高度一致。通过正反向设计特定折叠模式，能够实现精准的药物释放，具有高度可控性和灵活性。这一技术可应用于多种疾病如哮喘和糖尿病的治疗，有望实现复杂生理环境下的精准治疗（如图2）。</p> <p style="text-indent: 2em;
