Research on Nepalese medicinal plants are still limited to ethnopharmacological studies and qualitative phytochemical screening with a very few studies exploring their biological activities. This study aims to investigate biological activities of these plants and identify bioactive compounds present in each extract. A phytochemical profile of methanolic extracts of selected medicinal plants was established using high resolution (HR)-LCMS. Antioxidant activities were determined using DPPH, ABTS and FRAP assays. Highest DPPH radical scavenging was shown by Padamchal (IC50 = 3.47 ± 0.09), ABTS radical were most efficiently quenched by Pashanbed (IC50 = 3.82 ± 0.63) and the highest reducing potential was shown by Nirbikhi (FRAP = 61.76 ± 2.29 equivalent µg Fe2+/ml). In this study, phenolic compounds, in vitro anti-oxidant activity and anti-microbial properties have been investigated in five Himalayan medicinal plants, (e.g., Acorus calamus, Habe-naria intermedia, Hedychium spicatum, Roscoea procera and Valeriana jatamansi) in different solvent systems. R. procera exhibited significantly (p \ 0.05) higher phenolics; while H. spicatum was rich in flavonoids and V. jatamansi in anti-oxidant activity. Also, R. procera and H. spicatum were found rich in gallic acid; V. jatamansi in catechin, hydrox-ylbenzoic acid and caffeic acid and H. intermedia in hydroxyl benzoic acid. Solvent systems showed species specific response for extraction of total flavonoids and anti-oxidant activity. All the extracts were found effective against different bacterial and fungal strains in a dose dependent manner and maximum antimicrobial activity was found in R. procera as compared to other species. All the plant extracts showed greater activity against bacterial strains as compared to fungal strains. The aim of the present study was to investigate the phytochemical constituents (flavonoids and phenolics), antioxidant potential, cytotoxicity against HepG2 (human hepato carcinoma) cancer cell lines, and the antimicrobial activity of the methanol extract of selected traditional medicinal plants collected from Mizoram, India. A number of phenolic compounds were detected using HPLC-DAD-ESI-TOF-MS, mainly Luteolin, Kaempferol, Myricetin, Gallic Acid, Quercetin and Rutin, some of which have been described for the first time in the selected plants. The total phenolic and flavonoid contents showed high variation ranging from 4.44 to 181.91 µg of Gallic Acid equivalent per milligram DW (GAE/mg DW) and 3.17 to 102.2 µg of Quercetin/mg, respectively. The antioxidant capacity was determined by DPPH (IC 50 values ranges from 34.22 to 131.4 µg/mL), ABTS (IC 50 values ranges from 24.08 to 513.4 µg/mL), and reducing power assays. Antimicrobial activity was assayed against gram positive (Staphylococcus aureus), gram negative (Escherichia coli, Pseudomonas aeruginosa), and yeast (Candida albicans) demonstrating that the methanol extracts of some plants were efficacious antimicrobial agents. Additionally, cytotoxicity was assessed on human hepato carcinoma (HepG2) cancer cell lines and found that the extracts of Albizia lebbeck, Dillenia indica, and Bombax ceiba significantly decreased the cell viability at low concentrations with IC 50 values of 24.03, 25.09, and 29.66 µg/mL, respectively. The present study was undertaken to make a comparative analysis of antimicrobial and antioxidant properties of 8 medicinal plants, which are used in traditional medicine system. Methanolic extracts of Centella asiatica (leaves), Mentha piperita (leaves), Calotropis gigantean (leaves), Vitex nigundo (leaves), Bauhinia racemosus (leaves), Emblica officinalis (fruit), Bauhinia purpurea (leaves), Asperagus racemosus (stem) were tested for anti-microbial effects using the standard Agar Diffusion method. Antioxidant attributes of above extracts were studied by testing their ability to scavenge DPPH and ABTS radicles. Method Collected plant species were subjected to maceration in methanol for 72 hrs. Antioxidant activity of plant extracts was assessed by using DPPH free radical scavenging method in different concentrations (1 µg/ml, 3 µg/ml, 5 µg/ ml, 7 µg/ml and 10 µg/ml) and percentage inhibition and effective concentration (EC50) values were calculated. Result Result indicates that, EC50 of D. boryanum (3.75 µg/ml) and P. guajava (3.89 µg/ml) was less, EC50 of R. nepalensis (5.03 µg/ml) and S. japonica (6.75 µg/ml) was comparable and EC50 M. macrophylla (7.86 µg/ml), B. asiatica (9.14 µg/ml), E. adenophorum (7.78 µg/ml), E. crassipes (8.21 µg/ml), and N. arbortritis (8.16 µg/ml) was slightly higher than ascorbic acid (4.73 µg/ml). Conclusion Our result shows that, D. boryanum and P. gvajava possess higher antioxidant activity than the ascorbic acid implying that, they could be good free radical scavenging agents and should be EC50 = 4.12±1.27, ascorbic acid with EC50 = 6.20±1.67, gallic acid with EC50 = 4.75±1.24 and α-tocopherol with EC50 = 32.50±1.57 μg/mL. The experiment showed that aqueous fractions of the bark extracts of Abies pindrow (fraction: C2) and Cedrus deodara (fraction: E2) showed significantly lower EC50 values of 2.5±0.5 and 2.5±0.6 μg/mL, respectively. In reducing power assay, lower EC50 values of 5.5 and 4.5μg/mL were recorded for the aqueous fraction (fraction: C 2) and final residue (fraction: C5), of Abies pindrow, respectively. The ethyl acetate, acetone and final fractions of knot wood of Picea smithiana were found significantly active against all bacterial strains.
