It involves the addition of a methyl group to the cytosine residues in DNA, typically at CpG dinucleotides. This modification can silence <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/gene-expression-3169.html'>gene expression</a> by preventing the binding of transcription factors or by recruiting proteins that initiate <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/chromatin-2893.html'>chromatin</a> compaction [<a href="#1" title="1">1</a>]. DNA <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/methylation-4956.html'>methylation</a> is catalyzed by a family of enzymes called DNA Methyltransferases (DNMTs), which include DNMT1, DNMT3A, and DNMT3B.</p> <p>DNMT1 is primarily responsible for maintaining <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/methylation-4956.html'>methylation</a> patterns during DNA replication, while DNMT3A and DNMT3B are involved in de novo methylation.</p> <p><strong>Histone modifications: </strong>Histones are proteins around which DNA is wrapped to form nucleosomes. Histone modifications, such as acetylation, methylation, phosphorylation, and ubiquitination, can alter <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/chromatin-2893.html'>chromatin</a> structure and <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/gene-4901.html'>gene</a> expression. For example, histone acetylation generally promotes <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/gene-4901.html'>gene</a> transcription by relaxing chromatin, while histone deacetylation leads to <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/chromatin-2893.html'>chromatin</a> compaction and <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/gene-4901.html'>gene</a> silencing [<a href="#2" title="2">2</a>]. Histone modifications are regulated by a variety of enzymes, including Histone Acetyltransferases (HATs), Histone Deacetylases (HDACs), Histone Methyltransferases (HMTs), and Histone Demethylases (HDMs). These enzymes work in concert to establish and maintain specific patterns of histone modifications that influence <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/gene-4901.html'>gene</a> expression.</p> <p><strong>Non-coding RNAs:</strong> Non-coding RNAs, such as microRNAs (miRNAs) and long non-coding RNAs (lncRNAs), play crucial roles in regulating <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/gene-4901.html'>gene</a> expression. miRNAs are small RNA <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/molecules-6279.html'>molecules</a> that can bind to mRNAs and inhibit their translation or promote their degradation. lncRNAs are longer RNA <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/molecules-6279.html'>molecules</a> that can interact with chromatinmodifying complexes to regulate <a target='_blank' href='https://www.openaccessjournals.com/peer-reviewed-articles/gene-expression-3169.html'>gene expression</a> [<a href="#3" title="3">3</a>].
