We developed standard protocols for the isolation and cultivation for a range of marine and brackish microalgae. By comparing growth rates and lipid productivity, we assessed the potential of subtropical coastal and brackish microalgae for the production of biodiesel and other oil-based bioproducts. This study identified Nannochloropsis sp., Dunaniella salina and new isolates of Chlorella sp. and Tetraselmis sp. as suitable candidates for a multiple-product algae crop. We conclude that subtropical coastal microalgae display a variety of fatty acid profiles that offer a wide scope for several oil-based bioproducts, including biodiesel and omega-3 fatty acids. Biodiesel derived from microalgae is a potentially renewable and carbon-neutral alternative to petroleum fuels. One of the most important decisions in obtaining oil from microalgae is the choice of algal species to use. Eight microalgae from a total of 33 isolated cultures were selected based on their morphology and ease of cultivation. Five cultures were isolated from river and identified as strains of Scenedesmus obliquus YSR01, Nitzschia cf. pusilla YSR02, Chlorella ellipsoidea YSR03, S. obliquus YSR04, and S. obliquus YSR05, and three were isolated from wastewater and identified as S. obliquus YSW06, Micractinium pusillum YSW07, and Ourococcus multisporus YSW08, based on LSU rDNA (D1-D2) and ITS sequence analyses. S. obliquus YSR01 reached a growth rate of 1.68 ± 0.28 day À1 at 680 nm and a biomass concentration of 1.57 ± 0.67 g dwt L À1 , with a high lipid content of 58 ± 1.5%. Under similar environmental conditions, M. pusillum reached a growth rate of 2.3 ± 0.55 day À1 and a biomass concentration of 2.28 ± 0.16 g dwt L À1 , with a relatively low lipid content of 24 ± 0.5% w/w. The fatty acid compositions of the studied species were mainly myristic, palmitic, palmitoleic, oleic, linoleic, g-linolenic, and linolenic acids. Performance assessment of ten preselected microalgae under both batch and continuous culture points to the latter approach as the most adequate for evaluating fatty acid productivity. Differences were patent in continuous culture among strains that otherwise had analogous oil accumulation potential under batch culture. Some promising strains under batch culture (like Muriella aurantiaca and Monoraphidium braunii) exhibited, however, values for actual fatty acid productivity lower than 40 mg L-1 d-1 in continuous regime. The analysis performed in photochemostat under continuous culture regime revealed the great potential of Chlorococcum olefaciens, Pseudokirchneriella subcapitata and Scenedesmus almeriensis as oil producing microalgae. Fatty acid productivity levels over 90 mg L-1 d-1 were recorded for the latter strains under moderate nitrogen limitation, conditions which led to an enrichment in saturated and monounsaturated fatty acids, a more suitable profile as raw material for biodiesel. However, extracting and purifying oil from algae continues to prove a significant challenge in producing both microalgae byproducts and biofuel, as microbial oil extraction is relatively energy-intensive and costly. The aim of the research work is to produce biodiesel from micro algal species. Chaetoceros sp were identified for the research work and studied for their lipid, carbohydrate and protein content. The main aim of the project is to make use of the algae present in the water bodies and to extract the useful algal oil meant for biodiesel production to meet the challenges of fuel requirement in the present scenario. Microalgal oil was extracted from Chaetoceros sp. and the physicochemical properties were determined. The density, viscosity, acid value, saponification value and free fatty acids were recorded as 1.305gm/ml, 6.2mm 2 /s, 2.5339mg/gm of oil, 173.56mg/gm of oil, and 0.71gm/100 gm of algae (Oleic acid).
