The Interferon Gamma Release Assay (IGRA) is a blood test to find out if an individual is tuberculous positive or negative. This study applies statistical analysis to the clinical data of interferon-gamma levels of seventy-three subjects who diagnosed pulmonary TB in an anti-tuberculous treatment. Data analysis is performed to determine if there is a significant decline in interferon-gamma levels for the subjects during a period of six months, and to infer if the anti-tuberculous treatment is effective. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=data%20analysis" title="data analysis">data analysis</a>, <a href="https://publications.waset.org/abstracts/search?q=interferon%20gamma%20release%20assay" title=" interferon gamma release assay"> interferon gamma release assay</a>, <a href="https://publications.waset.org/abstracts/search?q=statistical%20methods" title=" statistical methods"> statistical methods</a>, <a href="https://publications.waset.org/abstracts/search?q=tuberculosis%20infection" title=" tuberculosis infection"> tuberculosis infection</a> </p> <a href="https://publications.waset.org/abstracts/49709/statistical-analysis-of-interferon-gh-for-the-effectiveness-of-an-anti-tuberculous-treatment" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/49709.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">305</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2804</span> Cloning, Expression and N-Terminal Pegylation of Human Interferon Alpha-2b Analogs and Their Cytotoxic Evaluation against Cancer Cell Lines</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Syeda%20Kiran%20Shahzadi">Syeda Kiran Shahzadi</a>, <a href="https://publications.waset.org/abstracts/search?q=Nasir%20Mahmood"> Nasir Mahmood</a>, <a href="https://publications.waset.org/abstracts/search?q=Muhammad%20Abdul%20Qadir"> Muhammad Abdul Qadir</a> </p> <p class="card-text"><strong>Abstract:</strong></p> In the current research, three recombinant human interferon alpha-2b proteins (two modified and one normal form) were produced and Pegylated with an aim to produce more effective drugs against viral infections and cancers. The modified recombinant human interferon alpha-2b proteins were produced by site-directed modifications of interferon alpha 2b gene, targeting the amino acids at positions ‘R23’ and ‘H34’. The resulting chemically modified and unmodified forms of human interferon alpha 2b were conjugated with methoxy-polyethylene glycol propanealdehyde (400 KDa) and methoxy-polyethylene glycol succinimidyl succinate (400 KDa). Pegylation of normal and modified forms of Interferon alpha-2b prolong their release time and enhance their efficacy. The conjugation of PEG with modified and unmodified human interferon alpha 2b protein drugs was also characterized with 1H-NMR, HPLC, and SDS-PAGE. Antiproliferative assays of modified and unmodified forms of drugs were performed in cell based bioassays using MDBK cell lines. The results indicated that experimentally produced recombinant human interferon alpha-2b proteins were biologically active and resulted in significant inhibition of cell growth. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=protein%20refolding" title="protein refolding">protein refolding</a>, <a href="https://publications.waset.org/abstracts/search?q=antiproliferative%20activities" title=" antiproliferative activities"> antiproliferative activities</a>, <a href="https://publications.waset.org/abstracts/search?q=biomedical%20applications" title=" biomedical applications"> biomedical applications</a>, <a href="https://publications.waset.org/abstracts/search?q=human%20interferon%20alpha-2b" title=" human interferon alpha-2b"> human interferon alpha-2b</a>, <a href="https://publications.waset.org/abstracts/search?q=pegylation" title=" pegylation"> pegylation</a>, <a href="https://publications.waset.org/abstracts/search?q=mPEG-propionaldehyde" title=" mPEG-propionaldehyde"> mPEG-propionaldehyde</a>, <a href="https://publications.waset.org/abstracts/search?q=site%20directed%20mutagenesis" title=" site directed mutagenesis"> site directed mutagenesis</a>, <a href="https://publications.waset.org/abstracts/search?q=E.%20coli%20expression" title=" E. coli expression"> E. coli expression</a> </p> <a href="https://publications.waset.org/abstracts/83956/cloning-expression-and-n-terminal-pegylation-of-human-interferon-alpha-2b-analogs-and-their-cytotoxic-evaluation-against-cancer-cell-lines" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/83956.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">177</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2803</span> Effect of Probiotics and Vitamin B on Plasma Interferon-Gamma and Interleukin-6 Levels in Active Pulmonary Tuberculosis </h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Yulistiani%20Yulistiani">Yulistiani Yulistiani</a>, <a href="https://publications.waset.org/abstracts/search?q=Zamrotul%20Izzah"> Zamrotul Izzah</a>, <a href="https://publications.waset.org/abstracts/search?q=Lintang%20Bismantara"> Lintang Bismantara</a>, <a href="https://publications.waset.org/abstracts/search?q=Wenny%20Putri%20Nilamsari"> Wenny Putri Nilamsari</a>, <a href="https://publications.waset.org/abstracts/search?q=Arif%20Bachtiar"> Arif Bachtiar</a>, <a href="https://publications.waset.org/abstracts/search?q=Budi%20Suprapti"> Budi Suprapti</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Interferon-gamma (IFN-γ) and interleukin-6 (IL-6) are pro-inflammatory cytokines, which have the protective immune response against Tuberculosis (TB). Indeed, pro-inflammatory cytokines Mycobacterium tuberculosis antigen-specific CD4+ and CD8+ T cells and NK cells increase the level of production of IFN-γ, a cytokine critical for augmenting the microbicidal activity of phagocytes. On the other hand, M. tuberculosis reduces the effects of IFN-γ by inhibiting the transcription of IFN-γ- responsive genes and by inducing the secretion of IL-6, which inhibits IFN-γ signaling. Probiotics Lactobacillus sp. and Bifidobacterium sp. were known to increase IFN-γ production in vivo, while vitamin B1, B6, and B12 worked on macrophages and releasing cytokines. Therefore, the present study was to evaluate the effect of probiotics and vitamin B supplement on changes of plasma cytokine levels in active pulmonary TB. From October to November 2016, twelve M. tuberculosis-infected patients starting anti-TB drugs were recruited, then divided into two groups. Seven patients were given a combination of probiotics and vitamin B, while five patients were in the control group. Plasma IFN-γ and IL-6 levels were measured by the ELISA kit before and a month after treatment. IFN-γ levels raised in four patients receiving the supplement (P = 0.743), while IL-6 increased in three patients in this group until day 30 of treatment (P = 0.298). Taken together, these results show the promising effect of probiotics and vitamin B on stimulation of IFN-γ and IL-6 production during intensive therapy of TB. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=interferon-gamma" title="interferon-gamma">interferon-gamma</a>, <a href="https://publications.waset.org/abstracts/search?q=interleukin-6" title=" interleukin-6"> interleukin-6</a>, <a href="https://publications.waset.org/abstracts/search?q=probiotic" title=" probiotic"> probiotic</a>, <a href="https://publications.waset.org/abstracts/search?q=tuberculosis" title=" tuberculosis"> tuberculosis</a> </p> <a href="https://publications.waset.org/abstracts/62781/effect-of-probiotics-and-vitamin-b-on-plasma-interferon-gamma-and-interleukin-6-levels-in-active-pulmonary-tuberculosis" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/62781.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">349</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2802</span> Pegylated Interferon in HCV Genotype 3 Relapser to Conventional Interferon in Pakistani Population</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Saad%20Khalid%20Niaz">Saad Khalid Niaz</a>, <a href="https://publications.waset.org/abstracts/search?q=Arif%20Mahmood%20Siddiqui"> Arif Mahmood Siddiqui</a>, <a href="https://publications.waset.org/abstracts/search?q=Afzal%20Haqi"> Afzal Haqi</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Background: Estimated prevalence of Hepatitis C in Pakistan is 5% of which 78 % are Genotype 3, in which Response to conventional interferon is reported to be 70%. Objective: To determine the efficacy of pegylated interferon 20 kDa (Unipeg) plus ribavirin (Ribazole) in HCV genotype 3 patients who relapsed to conventional interferon. Methods: This is an ongoing study of 20 enrolled patients. Pegylated interferon alfa-2a 20 kDa 180 mcg weekly with ribavirin, were administered for a period of 24 weeks. Virological Responses were measured by Qualitative HCV RNA at weeks 4, 12, 24 and 48 to determine Rapid Virological Response (RVR), Early Virological Response (EVR), End of Treatment (ETR) and Sustained Virological Response (SVR), respectively. EVR was done for those who didn’t achieve RVR. Results: Males were 12 (60%) and mean age was 38.5 ±7.62 years. Out of 20 recruited patients, all completed 4 weeks therapy; RVR was achieved in 8 (40%) patients. One patient was lost to follow up and one yet to visit at 12 weeks. From 10 patients, 8 (80%) patients achieved EVR. Out of intent-to-treat patients, 15 completed 24 weeks therapy, ETR was achieved in 14 (93%) patients and 9 patients completed post therapy follow-up, of which, 8 (89%) patients achieved SVR. Conclusion: Our interim data demonstrates that Pegylated Interferon alfa-2a 20 kDa 180 mcg (Unipeg) in combination with Ribavirin (Ribazole) has shown promising results in treating HCV Genotype 3 patients who relapsed to conventional interferon. We recommend use of Pegylated Interferon in Relapsers with Genotype 3 when financial constraints limit the use of oral antivirals. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=pegylated%20interferon%20%28unipeg%29" title="pegylated interferon (unipeg)">pegylated interferon (unipeg)</a>, <a href="https://publications.waset.org/abstracts/search?q=hepatitis%20c" title=" hepatitis c"> hepatitis c</a>, <a href="https://publications.waset.org/abstracts/search?q=relapsers" title=" relapsers"> relapsers</a>, <a href="https://publications.waset.org/abstracts/search?q=Pakistan" title=" Pakistan"> Pakistan</a> </p> <a href="https://publications.waset.org/abstracts/42845/pegylated-interferon-in-hcv-genotype-3-relapser-to-conventional-interferon-in-pakistani-population" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/42845.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">309</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2801</span> The Role of Vitamin D Supplementation in Augmenting IFN-γ Production in Response to Mycobacterium Tuberculosis Infection: A Randomized Controlled Trial</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Muhammad%20Imran%20Hussain">Muhammad Imran Hussain</a>, <a href="https://publications.waset.org/abstracts/search?q=Ramisha%20Ibtisam"> Ramisha Ibtisam</a>, <a href="https://publications.waset.org/abstracts/search?q=Tayyaba%20Fatima"> Tayyaba Fatima</a>, <a href="https://publications.waset.org/abstracts/search?q=Huba%20Khalid"> Huba Khalid</a>, <a href="https://publications.waset.org/abstracts/search?q=Ayesha%20Aziz"> Ayesha Aziz</a>, <a href="https://publications.waset.org/abstracts/search?q=Khansa"> Khansa</a>, <a href="https://publications.waset.org/abstracts/search?q=Adan%20Sitara"> Adan Sitara</a>, <a href="https://publications.waset.org/abstracts/search?q=Anam%20Shahzad"> Anam Shahzad</a>, <a href="https://publications.waset.org/abstracts/search?q=Aymen%20Jabeen"> Aymen Jabeen</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Vitamin D supports the immune system fight TB by inhibiting Interferon-gamma (IFN-γ) and lowering host inflammation. The purpose of the research was to see if giving the vitamin D supplements to TB patients affected their prognosis. A randomized placebo control study of 200 TB patients was performed among which 106 received 400,000 IU of injectable vitamin D3 and 94 received placebo for 2 doses. Assessment was carried out at the end of every month for 3 months. IFN-γ responses to whole blood stimulation generated by the Mycobacterium tuberculosis sonicate (MTBs) antigen and early secreted and T cell activated 6 kDa (ESAT6) were assessed at 0 and 12 weeks. The statistical analysis used descriptive statistics (mean and standard deviation), Friedman's test and Fisher's test. The vitamin D group gained significantly more weight (+3.90 pounds) and had less persistent lung disease on imaging (1.33 zones vs. 1.84 zones). They also had a 50% decrease in cavity size. Additionally, patients with low baseline serum concentrations of 25-(OH)D had a significant increase in MTB-induced IFN-γ production after taking vitamin D supplements. Vitamin D administration in large amounts can hasten the recovery of TB patients. The findings point is a therapeutically useful activity of Vitamin D's in the management for tuberculosis. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=tuberculosis" title="tuberculosis">tuberculosis</a>, <a href="https://publications.waset.org/abstracts/search?q=vitamin%20D" title=" vitamin D"> vitamin D</a>, <a href="https://publications.waset.org/abstracts/search?q=interferon%20gamma" title=" interferon gamma"> interferon gamma</a>, <a href="https://publications.waset.org/abstracts/search?q=protein" title=" protein"> protein</a>, <a href="https://publications.waset.org/abstracts/search?q=infection" title=" infection"> infection</a> </p> <a href="https://publications.waset.org/abstracts/184471/the-role-of-vitamin-d-supplementation-in-augmenting-ifn-gh-production-in-response-to-mycobacterium-tuberculosis-infection-a-randomized-controlled-trial" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/184471.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">52</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2800</span> Evaluation of the Synergistic Inhibition of Enterovirus 71 Infection by Interferon-α Coupled with Pleconaril in RD Cells</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Wen-Yu%20Lin">Wen-Yu Lin</a>, <a href="https://publications.waset.org/abstracts/search?q=Yi-Ching%20Chung"> Yi-Ching Chung</a>, <a href="https://publications.waset.org/abstracts/search?q=Tzyy-Rong%20Jinn"> Tzyy-Rong Jinn</a> </p> <p class="card-text"><strong>Abstract:</strong></p> It is well known that enterovirus 71 (EV71) causes recurring outbreaks of hand, foot and mouth disease (HFMD) and encephalitis leading to complications or death in young children. And, several HFMD of EV71 with high mortalities occurred in Asia countries, such as Malaysia (1997), Taiwan (1998) and China (2008). Thus, more effective antiviral drugs are needed to prevent or reduce EV71-related complications. As reported, interferon-α protects mice from lethal EV71 challenge by the modulation of innate immunity and then degrade enterovirus protease 3Cᵖʳᵒ. On the other side, pleconaril by targeting enterovirus VP1 protein and then block virus entry and attachment. Thus, the aim of this study was to evaluate the synergistic antiviral activity of interferon-α and pleconaril against enterovirus 71 infection. In a preliminary study showed that pleconaril at concentrations of 50, 100 and 300 µg/mL reduced EV71-induced CPE to 52.0 ± 2.5%, 40.2 ± 3.5% and 26.5 ± 1.5%, respectively, of that of the EV71-infected RD control cells (taken as 100%). Notably, 1000 IU/mL of interferon-α in combination with pleconaril at concentrations of 50, 100 and 300µg/mL suppressed EV71-induced CPE by 30.2 ± 3.8%, 16.5 ± 1.3% and 2.8 ± 2.0%, respectively, of that of the pleconaril alone treated with the infected RD cells. These results indicated that interferon-α 1000 IU/mL combination with pleconaril (50, 100 and 300µg/mL) inhibited EV71-induced CPE more effectively than treated with pleconaril alone in the infected RD cells. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=enterovirus%2071" title="enterovirus 71">enterovirus 71</a>, <a href="https://publications.waset.org/abstracts/search?q=interferon-%CE%B1" title=" interferon-α"> interferon-α</a>, <a href="https://publications.waset.org/abstracts/search?q=pleconaril" title=" pleconaril"> pleconaril</a>, <a href="https://publications.waset.org/abstracts/search?q=RD%20cells" title=" RD cells"> RD cells</a> </p> <a href="https://publications.waset.org/abstracts/102058/evaluation-of-the-synergistic-inhibition-of-enterovirus-71-infection-by-interferon-a-coupled-with-pleconaril-in-rd-cells" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/102058.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">140</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2799</span> Effect of Gamma Radiation on Bromophenol Blue Dyed Films as Dosimeter</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Priyanka%20R.%20Oberoi">Priyanka R. Numerous medical devices are sterilized by ionizing radiation. In the current research paper, an attempt was made to develop precise and inexpensive polymeric film dosimeter which can be used for controlling radiation dosage. Polymeric film containing (pH sensitive dye) indicator dye Bromophenol blue (BPB) was casted to check the effect of Gamma radiation on its optical and physical properties. The film was exposed to gamma radiation at 4 kGy/hr in the range of 0 to 300 kGy at an interval of 50 kGy. Release of vinyl acetate from an emulsion on high radiation reacts with the BPB fading the color of the film from blue to light blue and then finally colorless, indicating a change in pH from basic to acidic form. The change was characterized by using CIE l*a*b*, ultra-violet spectroscopy and FT-IR respectively. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=bromophenol%20blue" title="bromophenol blue">bromophenol blue</a>, <a href="https://publications.waset.org/abstracts/search?q=dosimeter" title=" dosimeter"> dosimeter</a>, <a href="https://publications.waset.org/abstracts/search?q=gamma%20radiation" title=" gamma radiation"> gamma radiation</a>, <a href="https://publications.waset.org/abstracts/search?q=polymer" title=" polymer"> polymer</a> </p> <a href="https://publications.waset.org/abstracts/55840/effect-of-gamma-radiation-on-bromophenol-blue-dyed-films-as-dosimeter" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/55840.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">290</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2798</span> Development, Optimization and Characterization of Gastroretentive Multiparticulate Drug Delivery System</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Swapnila%20V.%20Vanshiv">Swapnila V. The formulation involved hollow microsphere preparation by using solvent evaporation technique. Concentrations of rate controlling polymer, hydrophilic polymer, internal phase ratio, stirring speed were optimized to get desired responses, namely release of Dipyridamole, buoyancy of microspheres, entrapment efficiency of microspheres. In the formulation, the floating microspheres were prepared by using ethyl cellulose as release retardant and HPMC as a low density hydrophilic swellable polymer. Formulated microspheres were evaluated for their physical properties such as particle size and surface morphology by optical microscopy and SEM. Entrapment efficiency, floating behavior and drug release study as well the formulation was evaluated for in vivo gastroretention in rabbits using gamma scintigraphy. Formulation showed 75% drug release up to 10 hr with entrapment efficiency of 91% and 88% buoyancy till 10 hr. Gamma scintigraphic studies revealed that the optimized system was retained in the gastric region (stomach) for a prolonged period i.e. more than 5 hr. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=Dipyridamole%20microspheres" title="Dipyridamole microspheres">Dipyridamole microspheres</a>, <a href="https://publications.waset.org/abstracts/search?q=gastroretention" title=" gastroretention"> gastroretention</a>, <a href="https://publications.waset.org/abstracts/search?q=HPMC" title=" HPMC"> HPMC</a>, <a href="https://publications.waset.org/abstracts/search?q=optimization%20method" title=" optimization method"> optimization method</a> </p> <a href="https://publications.waset.org/abstracts/8657/development-optimization-and-characterization-of-gastroretentive-multiparticulate-drug-delivery-system" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/8657.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">385</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2797</span> Inf-γ and Il-2 Asses the Therapeutic Response in Anti-tuberculosis Patients at Jamot Hospital Yaounde, Cameroon</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Alexandra%20Emmanuelle%20Membangbi">Alexandra Emmanuelle Membangbi</a>, <a href="https://publications.waset.org/abstracts/search?q=Jacky%20Njiki%20Biko%C3%AF"> Jacky Njiki Bikoï</a>, <a href="https://publications.waset.org/abstracts/search?q=Esther%20Del-florence%20Moni%20Ndedi"> Esther Del-florence Moni Ndedi</a>, <a href="https://publications.waset.org/abstracts/search?q=Marie%20Joseph%20Nkodo%20Mindimi"> Marie Joseph Nkodo Mindimi</a>, <a href="https://publications.waset.org/abstracts/search?q=Donatien%20Serge%20Mbaga"> Donatien Serge Mbaga</a>, <a href="https://publications.waset.org/abstracts/search?q=Elsa%20Nguiffo%20Makue"> Elsa Nguiffo Makue</a>, <a href="https://publications.waset.org/abstracts/search?q=Andr%C3%A9%20Chris%20Mikangue%20Mbongue"> André Chris Mikangue Mbongue</a>, <a href="https://publications.waset.org/abstracts/search?q=Martha%20Mesembe"> Martha Mesembe</a>, <a href="https://publications.waset.org/abstracts/search?q=George%20Ikomey%20Mondinde"> George Ikomey Mondinde</a>, <a href="https://publications.waset.org/abstracts/search?q=Eric%20Walter%20Perfura-yone"> Eric Walter Perfura-yone</a>, <a href="https://publications.waset.org/abstracts/search?q=Sara%20Honorine%20Riwom%20Essama"> Sara Honorine Riwom Essama</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Background: Tuberculosis (TB) is one of the top lethal infectious diseases worldwide. In recent years, interferon-γ (INF-γ) release assays (IGRAs) have been established as routine tests for diagnosing TB infection. However, produced INF-γ assessment failed to distinguish active TB (ATB) from latent TB infection (LTBI), especially in TB epidemic areas. In addition to IFN-γ, interleukin-2 (IL-2), another cytokine secreted by activated T cells, is also involved in immune response against Mycobacterium tuberculosis. The aim of the study was to assess the capacity of IFN-γ and IL2 to evaluate the therapeutic response of patients on anti-tuberculosis treatment. Material and Methods: We conducted a cross-sectional study in the Pneumonology Departments of the Jamot Hospital in Yaoundé between May and August 2021. After signed the informed consent, the sociodemographic data, as well as 5 mL of blood, were collected in the crook of the elbow of each participant. Sixty-one subjects were selected (n= 61) and divided into 4 groups as followed: group 1: resistant tuberculosis (n=13), group 2: active tuberculosis (n=19), group 3 cured tuberculosis (n=16), and group 4: presumed healthy persons (n=13). The cytokines of interest were determined using an indirect Enzyme-linked Immuno-Sorbent Assay (ELISA) according to the manufacturer's recommendations. P-values < 0.05 were interpreted as statistically significant. All statistical calculations were performed using SPSS version 22.0 Results: The results showed that men were more 14/61 infected (31,8%) with a high presence in active and resistant TB groups. The mean age was 41.3±13.1 years with a 95% CI = [38.2-44.7], the age group with the highest infection rate was ranged between 31 and 40 years. The IL-2 and INF-γ means were respectively 327.6±160.6 pg/mL and 26.6±13.0 pg/mL in active tuberculosis patients, 251.1±30.9 pg/mL and 21.4±9.2 pg/mL in patients with resistant tuberculosis, while it was 149.3±93.3 pg/mL and 17.9±9.4 pg/mL in cured patients, 15.1±8.4 pg/mL and 5.3±2.6 pg/mL in participants presumed healthy (p <0.0001). Significant differences in IFN-γ and IL-2 rates were observed between the different groups. Conclusion: Monitoring the serum levels of INF-γ and IL-2 would be useful to evaluate the therapeutic response of anti-tuberculosis patients, particularly in the both cytokines association case, that could improve the accuracy of routine examinations. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=antibiotic%20therapy" title="antibiotic therapy">antibiotic therapy</a>, <a href="https://publications.waset.org/abstracts/search?q=interferon%20gamma" title=" interferon gamma"> interferon gamma</a>, <a href="https://publications.waset.org/abstracts/search?q=interleukin%202" title=" interleukin 2"> interleukin 2</a>, <a href="https://publications.waset.org/abstracts/search?q=tuberculosis" title=" tuberculosis"> tuberculosis</a> </p> <a href="https://publications.waset.org/abstracts/161902/inf-gh-and-il-2-asses-the-therapeutic-response-in-anti-tuberculosis-patients-at-jamot-hospital-yaounde-cameroon" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/161902.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">117</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2796</span> A Systematic Review for the Association between Active Smoking and Latent Tuberculosis Infection</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Pui%20Hong%20%20Chung">Pui Hong Chung</a>, <a href="https://publications.waset.org/abstracts/search?q=Wing%20Chi%20Ho"> Wing Chi Ho</a>, <a href="https://publications.waset.org/abstracts/search?q=Jun%20Li"> Jun Li</a>, <a href="https://publications.waset.org/abstracts/search?q=Cyrus%20%20Leung"> Cyrus Leung</a>, <a href="https://publications.waset.org/abstracts/search?q=Ek%20Yeoh"> Ek Yeoh</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Background: Cigarette smoking is associated with poor tuberculosis (TB) outcomes in terms of progression of active TB, relapse of TB and TB-related mortality, but the association with latent tuberculosis infection (LTBI) is unclear. The systematic review conducted aimed at studying the association between active smoking and LTBI, and likelihood of dose-response relationship. Methods: Two independent reviewers searched three electronic databases comprising PudMed, Medline by EBSCOHOST, ExcerptaMedica Database (EMBASE), from inception up to 31st Dec 2015 for studies reporting data on current smoking and the LTBI with tuberculin skin test (TST) or interferon-γ release assays (IGRAs) results, comparing the odds ratios (ORs) of outcome measure of TST or IGRAs among current smokers with 95% confidence intervals (CI). Results: Seven studies were identified, including six cross-sectional studies and one longitudinal cohort study. The outcome measures from three studies were in TST, three studies in IGRAs and one for both tests. For TST, OR ranging from 1.39 to 3.40 (95% CI) with all studies shown positive association between cigarette smoking and LTBI. For IGRAs, OR ranging from 0.47 to 1.89 (95% CI) with one study shown the negative association that might be related to impaired interferon-gamma production in immunosuppressive persons. One identified study demonstrated positive dose-response relationship in TST result. Conclusions: Cigarette smoking is likely to be a risk factor of LTBI. There is the important implication for TB and tobacco control program to halt TB by empowering public health policy. Further study is also needed to provide more evidence of the dose-response model/relationship. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=latent%20tuberculosis%20infection" title="latent tuberculosis infection">latent tuberculosis infection</a>, <a href="https://publications.waset.org/abstracts/search?q=systematic%20review" title=" systematic review"> systematic review</a>, <a href="https://publications.waset.org/abstracts/search?q=active%20smoking" title=" active smoking"> active smoking</a>, <a href="https://publications.waset.org/abstracts/search?q=model" title=" model"> model</a> </p> <a href="https://publications.waset.org/abstracts/59424/a-systematic-review-for-the-association-between-active-smoking-and-latent-tuberculosis-infection" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/59424.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">256</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2795</span> Impact of Weather Conditions on Generalized Frequency Division Multiplexing over Gamma Gamma Channel</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Muhammad%20Sameer%20Ahmed">Muhammad Sameer Ahmed</a>, <a href="https://publications.waset.org/abstracts/search?q=Piotr%20Remlein"> Piotr Remlein</a>, <a href="https://publications.waset.org/abstracts/search?q=Tansal%20Gucluoglu"> Tansal Gucluoglu</a> </p> <p class="card-text"><strong>Abstract:</strong></p> The technique called as Generalized frequency division multiplexing (GFDM) used in the free space optical channel can be a good option for implementation free space optical communication systems. NaI (Tl) scintillation detector is the most commonly used in radiation and medical physics for measurement of the gamma ray activity of various samples. In addition, the scintillation detector has a lot of applications in the elemental analysis of various compounds, alloys using activation analysis. In each application for quantitative analysis, it is very much essential to know the detection efficiency and resolution for different gamma energies. In this work, the energy dependence of efficiency and resolution of NaI (Tl) detector using gamma-ray spectroscopy are investigated. Different photon energies of 356.01 keV,511keV,661.60keV,1170 keV,1274.53 keV and 1330 keV are obtained from four radioactive sources (133Ba,22Na,137Cs and 60 Co) used in these studies. Values of full energy peak efficiencies of these gamma energies are found to be respectively 58.46%,10.15%,14.39%,1.4%,3.27% and 1.31%. The values of percent resolution for above different gamma ray energies are found to be 11.27%,7.27%,6.38%,5.17%,4.86% and 4.74% respectively. It was found that the efficiency of the detector exponentially decreases with energy and the resolution of the detector is directly proportional to the energy of gamma-ray. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=naI%20%28Tl%29%20gamma-ray%20spectrometer" title="naI (Tl) gamma-ray spectrometer">naI (Tl) gamma-ray spectrometer</a>, <a href="https://publications.waset.org/abstracts/search?q=resolution" title=" resolution"> resolution</a>, <a href="https://publications.waset.org/abstracts/search?q=full%20energy%20peak%20efficiency" title=" full energy peak efficiency"> full energy peak efficiency</a>, <a href="https://publications.waset.org/abstracts/search?q=radioactive%20sources" title=" radioactive sources"> radioactive sources</a> </p> <a href="https://publications.waset.org/abstracts/158239/determination-of-full-energy-peak-efficiency-and-resolution-of-nai-tl-detector-using-gamma-ray-spectroscopy" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/158239.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">104</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2792</span> Successful Treatment of Multifocal XDR Tuberculosis Osteomyelitis</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Abeer%20N.%20Alshukairi">Abeer N. The major advantages are its ability to kill radio-resistant tumor and its anti-metastatic activity. Poly(ADP-ribose) polymerase-1 (PARP-1) inhibitors have been widely used as radiosensitizer, but its role in metastasis is unknown. The purpose of our study was to investigate the effect of PARP-1 depletion in combination with either Carbon Ion Beam (CIB) or gamma irradiation on metastatic potential of cultured cancerous cells. A549 cells were irradiated with CIB (0-4Gy) or gamma (0, 2, 4, 6 and 10 Gy) with and without PARP-1 inhibition. The metastatic potential of the cells was determined by cell migratory assay, expression, and activity of MMP-2 and MMP-9, expression of Cadherin, Fibronectin, and Vimentin. CIB exposure reduced migratory property and activity of MMP-2 and MMP-9 significantly. CIB with PARP-1 inhibition reduced cell migration and Matrix Metalloproteinase (MMPs) activity in a synergistic manner. Expression of MMPs was also down-regulated in CIB and combined treatment. On the contrary, MMP- 2 and MMP-9 activity was significantly increased in gamma irradiated cells but decreased upon combined treatment of gamma and PARP-1 inhibitor. MMPs expression and migration was reduced when gamma irradiation was combined with PARP-1 inhibition. Thus, our study clearly demonstrates that PARP-1 inhibition in combination with either high or low LET can significantly suppress metastatic potential in cancer cells and thereby can be a promising tool in controlling metastatic cancers. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=high%20LET" title="high LET">high LET</a>, <a href="https://publications.waset.org/abstracts/search?q=low%20LET" title=" low LET"> low LET</a>, <a href="https://publications.waset.org/abstracts/search?q=matrix%20metalloproteinase%20%28MMP%29" title=" matrix metalloproteinase (MMP)"> matrix metalloproteinase (MMP)</a>, <a href="https://publications.waset.org/abstracts/search?q=PARP-1" title=" PARP-1"> PARP-1</a> </p> <a href="https://publications.waset.org/abstracts/76226/combined-treatment-of-parp-1-inhibitor-and-carbon-ion-or-gamma-exposure-reduces-the-metastatic-potential-in-cultured-human-cells" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/76226.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">214</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2790</span> Gamma Irradiation Effect on Structural and Optical Properties of Bismuth-Boro-Tellurite Glasses</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Azuraida%20Amat">Azuraida Amat</a>, <a href="https://publications.waset.org/abstracts/search?q=Halimah%20Mohamed%20Kamari"> Halimah Mohamed Kamari</a>, <a href="https://publications.waset.org/abstracts/search?q=Che%20Azurahanim%20Che%20Abdullah"> Che Azurahanim Che Abdullah</a>, <a href="https://publications.waset.org/abstracts/search?q=Ishak%20Mansor"> Ishak Mansor</a> </p> <p class="card-text"><strong>Abstract:</strong></p> The changes of the optical and structural properties of Bismuth-Boro-Tellurite glasses pre and post gamma irradiation were studied. The Fourier Transform Infrared Spectroscopy (FTIR) was used to explore the structural bonding in the prepared glass samples due to exposure, while UV-VIS Spectrophotometer was used to evaluate the changes in the optical properties before and after irradiation. Gamma irradiation causes a profound changes in the peak intensity as shown by FTIR spectra which is due to the breaking of the network bonding. Before gamma irradiation, the optical band gap, Eg value decreased from 2.44 eV to 2.15 eV with the addition of Bismuth content. The value kept decreasing (from 2.18 eV to 2.00 eV) following exposure to gamma radiation due to the increase of non-bridging oxygen (NBO) and the increase of defects in the glass. In conclusion, the glass with high content of Bi2O3 (0.30Bi) give the smallest Eg and show less changes in FTIR spectra after gamma irradiation, which indicate that this glass is more resistant to gamma radiation compared to other glasses. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=boro-tellurite" title="boro-tellurite">boro-tellurite</a>, <a href="https://publications.waset.org/abstracts/search?q=bismuth" title=" bismuth"> bismuth</a>, <a href="https://publications.waset.org/abstracts/search?q=gamma%20radiation" title=" gamma radiation"> gamma radiation</a>, <a href="https://publications.waset.org/abstracts/search?q=optical%20properties" title=" optical properties"> optical properties</a> </p> <a href="https://publications.waset.org/abstracts/25457/gamma-irradiation-effect-on-structural-and-optical-properties-of-bismuth-boro-tellurite-glasses" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/25457.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">427</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2789</span> Citrullinated Myelin Basic Protein Mediated Inflammation in Astrocytes</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Lali%20Shanshiashvili">Lali Shanshiashvili</a>, <a href="https://publications.waset.org/abstracts/search?q=Marika%20Chikviladze"> Marika Chikviladze</a>, <a href="https://publications.waset.org/abstracts/search?q=Nino%20Mamulashvili"> Nino Mamulashvili</a>, <a href="https://publications.waset.org/abstracts/search?q=Maia%20Sepashvili"> Maia Sepashvili</a>, <a href="https://publications.waset.org/abstracts/search?q=Nana%20Narmania"> Nana Narmania</a>, <a href="https://publications.waset.org/abstracts/search?q=David%20Mikeladze"> David Mikeladze</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Purpose: During demyelinating inflammatory diseases and after the damage of the myelin sheet, myelin-derived proteins, including myelin basic protein (MBP), are secreted into the extracellular space. MBP shows extensive post-translational modifications, including the deimination of arginine residues. Deiminated MBP is structurally less ordered, susceptible to proteolytic attack, and more immunogenic than the unmodified one. It is hypothesized that MBP could change the inflammatory response in astrocytes. Methods: MBP was isolated and purified from bovine brain white matter. Primary astrocyte cultures were prepared from whole brains of 2-day-old Wistar rats. For evaluation of glutamate uptake/release in astrocytes following treatment of cells with MBP charge isomers, Glutamate Assay Kit was used. The expression of EAAT-2 (excitatory amino acid transporters), peroxisome proliferator-activated receptor gamma (PPAR- γ), inhibitor of nuclear factor kappa B (IkB), and high mobility group protein B1 (HMGB1) in astrocytes were assayed by Western Blot analysis. Results: This study investigated the action of deiminated isomer (C8) on the cultured primary astrocytes and compared its effects with the effects of unmodified C1 isomers. The study found that C8 and C1 MBP differently act on the uptake and release of glutamate in astrocytes: nonmodified C1 MBP increases the uptake of glutamate and does not change the release, whereas C8 decreases the release of glutamate but does not alter the uptake. Nevertheless, both isomers increased the expression of PPAR-γ and EAAT2 in the same intensity. However, immunostaining and Western Blots of cell lysates showed a decrease of IkB and increased expression of HMGB1 after the treatment of astrocytes by C8. Moreover, in the presence of C8, astrocytes release more nitric oxide than unmodified C1 isomers. Conclusion: These data suggest that the deiminated isomer of MBP evokes an inflammatory response and enhances the ability of astrocytes to release proinflammatory mediators through activation of NF-kB after the breakdown of myelin sheets. Atenolol shows pH dependent solubility resulting into a bioavailability of 36%. Thus, site specific oral controlled release floating drug delivery system was developed. Formulation includes novice use of rate controlling polymer such as locust bean gum (LBG) in combination of HPMC K4M and gas generating agent sodium bicarbonate. Tablet was prepared by direct compression method and evaluated for physico-mechanical properties. The statistical method was utilized to optimize the effect of independent variables, namely amount of HPMC K4M, LBG and three dependent responses such as cumulative drug release, floating lag time, floating time. Graphical and mathematical analysis of the results allowed the identification and quantification of the formulation variables influencing the selected responses. To study the gastrointestinal transit of the optimized gastro-retentive formulation, in vivo gamma scintigraphy was carried out in six healthy rabbits, after radio labeling the formulation with 99mTc. The transit profiles demonstrated that the dosage form was retained in the stomach for more than 5 hrs. The study signifies the potential of the developed system for stomach targeted delivery of atenolol with improved bioavailability. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=floating%20tablet" title="floating tablet">floating tablet</a>, <a href="https://publications.waset.org/abstracts/search?q=factorial%20design" title=" factorial design"> factorial design</a>, <a href="https://publications.waset.org/abstracts/search?q=gamma%20scintigraphy" title=" gamma scintigraphy"> gamma scintigraphy</a>, <a href="https://publications.waset.org/abstracts/search?q=antihypertensive%20model%20drug" title=" antihypertensive model drug"> antihypertensive model drug</a>, <a href="https://publications.waset.org/abstracts/search?q=HPMC" title=" HPMC"> HPMC</a>, <a href="https://publications.waset.org/abstracts/search?q=locust%20bean%20gum" title=" locust bean gum"> locust bean gum</a> </p> <a href="https://publications.waset.org/abstracts/8593/optimization-of-gastro-retentive-matrix-formulation-and-its-gamma-scintigraphic-evaluation" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/8593.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">275</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2787</span> Biological Studies on Producing Samoli Bread Supplement with Irradiated Sunflower Flour by Gamma Rays</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Amal.%20N.%20Al-Kuraieef">Amal. In the present study Polyphenon 60 (P60) and ciprofloxacin (Cipro) were loaded in a single nano emulsion (NE) system via ultra-sonication technique and characterized for particle size, in vitro release and antibacterial efficacy against Bcl-2 level Escherichia coli bacteria. To determine in vivo pharmacokinetic parameters and intravaginal transportation of NE, gamma scintigraphy and biodistribution study was conducted by radiolabelling NE with technetium pertechnetate (99mTc). The preliminary antibacterial investigation showed synergy between these compounds with FICindex of 0.42. The developed formulation showed zeta potential +55.3 and particle size of 151.7 nm, with PDI of 0.196. The in vitro release percentage of P60 at the end of 7th hours was 94.8 ± 0.9 % whereas the release for Cipro was 75.1± 0.15 % in simulated vaginal media. MBC was identified and the findings demonstrated that in both ESBL (Extended Spectrum β- lactamase) and MBL (Metallo β- lactamase) cultures the P60+Cipro NE showed inhibition of growth of all the isolates at 2 mg/ml dilutions. The percentage per gram of radiolabelled drug was found (3.50±0.26) and (3.81±0.30) in kidney and urinary bladder, respectively at 3 h. From the findings, it was concluded that the developed P60+Cipro NE was transported efficiently throughout the target organs, had long duration of action and high biocompatibility via intravaginal administration as compared to oral administration. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=ciprofloxacin" title="ciprofloxacin">ciprofloxacin</a>, <a href="https://publications.waset.org/abstracts/search?q=gamma%20scintigraphy" title=" gamma scintigraphy"> gamma scintigraphy</a>, <a href="https://publications.waset.org/abstracts/search?q=intravaginal%20drug%20delivery" title=" intravaginal drug delivery"> intravaginal drug delivery</a>, <a href="https://publications.waset.org/abstracts/search?q=Polyphenon%2060" title=" Polyphenon 60"> Polyphenon 60</a> </p> <a href="https://publications.waset.org/abstracts/59387/antimicrobial-evaluation-of-polyphenon-60-and-ciprofloxacin-loaded-nano-emulsion-against-uropathogenic-escherichia-coli-bacteria-and-its-in-vivo-analysis" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/59387.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">319</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2784</span> Effect of Radioprotectors on DNA Repair Enzyme and Survival of Gamma-Irradiated Cell Division Cycle Mutants of Saccharomyces pombe</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Purva%20Nemavarkar">Purva Nemavarkar</a>, <a href="https://publications.waset.org/abstracts/search?q=Badri%20Narain%20Pandey"> Badri Narain Pandey</a>, <a href="https://publications.waset.org/abstracts/search?q=Jitendra%20Kumar"> Jitendra Kumar</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Introduction: The objective was to understand the effect of various radioprotectors on DNA damage repair enzyme and survival in gamma-irradiated wild and cdc mutants of S. pombe (fission yeast) cultured under permissive and restrictive conditions. DNA repair process, as influenced by radioprotectors, was measured by activity of DNA polymerase in the cells. The use of single cell gel electrophoresis assay (SCGE) or Comet Assay to follow gamma-irradiation induced DNA damage and effect of radioprotectors was employed. In addition, studying the effect of caffeine at different concentrations on S-phase of cell cycle was also delineated. Materials and Methods: S. pombe cells grown at permissive temperature (250C) and/or restrictive temperature (360C) were followed by gamma-radiation. Percentage survival and activity of DNA Polymerase (yPol II) were determined after post-irradiation incubation (5 h) with radioprotectors such as Caffeine, Curcumin, Disulphiram, and Ellagic acid (the dose depending on individual D 37 values). The gamma-irradiated yeast cells (with and without the radioprotectors) were spheroplasted by enzyme glusulase and subjected to electrophoresis. Radio-resistant cells were obtained by arresting cells in S-phase using transient treatment of hydroxyurea (HU) and studying the effect of caffeine at different concentrations on S-phase of cell cycle. Results: The mutants of S. pombe showed insignificant difference in survival when grown under permissive conditions. However, growth of these cells under restrictive temperature leads to arrest in specific phases of cell cycle in different cdc mutants (cdc10: G1 arrest, cdc22: early S arrest, cdc17: late S arrest, cdc25: G2 arrest). All the cdc mutants showed decrease in survival after gamma radiation when grown at permissive and restrictive temperatures. Inclusion of the radioprotectors at respective concentrations during post irradiation incubation showed increase in survival of cells. Activity of DNA polymerase enzyme (yPol II) was increased significantly in cdc mutant cells exposed to gamma-radiation. Following SCGE, a linear relationship was observed between doses of irradiation and the tail moments of comets. The radioprotection of the fission yeast by radioprotectors can be seen by the reduced tail moments of the yeast comets. Caffeine also exhibited its radio-protective ability in radio-resistant S-phase cells obtained after HU treatment. Conclusions: The radioprotectors offered notable radioprotection in cdc mutants when added during irradiation. The present study showed activation of DNA damage repair enzyme (yPol II) and an increase in survival after treatment of radioprotectors in gamma irradiated wild type and cdc mutants of S. pombe cells. Results presented here showed feasibility of applying SCGE in fission yeast to follow DNA damage and radioprotection at high doses, which are not feasible with other eukaryotes. Inclusion of caffeine at 1mM concentration to S phase cells offered protection and did not decrease the cell viability. It can be proved that at minimal concentration, caffeine offered marked radioprotection. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=radiation%20protection" title="radiation protection">radiation protection</a>, <a href="https://publications.waset.org/abstracts/search?q=cell%20cycle" title=" cell cycle"> cell cycle</a>, <a href="https://publications.waset.org/abstracts/search?q=fission%20yeast" title=" fission yeast"> fission yeast</a>, <a href="https://publications.waset.org/abstracts/search?q=comet%20assay" title=" comet assay"> comet assay</a>, <a href="https://publications.waset.org/abstracts/search?q=s-phase" title=" s-phase"> s-phase</a>, <a href="https://publications.waset.org/abstracts/search?q=DNA%20repair" title=" DNA repair"> DNA repair</a>, <a href="https://publications.waset.org/abstracts/search?q=radioprotectors" title=" radioprotectors"> radioprotectors</a>, <a href="https://publications.waset.org/abstracts/search?q=caffeine" title=" caffeine"> caffeine</a>, <a href="https://publications.waset.org/abstracts/search?q=curcumin" title=" curcumin"> curcumin</a>, <a href="https://publications.waset.org/abstracts/search?q=SCGE" title=" SCGE"> SCGE</a> </p> <a href="https://publications.waset.org/abstracts/164879/effect-of-radioprotectors-on-dna-repair-enzyme-and-survival-of-gamma-irradiated-cell-division-cycle-mutants-of-saccharomyces-pombe" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/164879.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">113</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2783</span> Effects of Gamma Irradiation on Chemical and Antioxidant Properties of Iranian Native Fresh Barberry Fruit</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Samira%20Berenji%20Ardestani">Samira Berenji Ardestani</a>, <a href="https://publications.waset.org/abstracts/search?q=Hamid%20Reza%20Akhavan"> Hamid Reza Akhavan</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Gamma irradiation greatly reduces the potential microbiological risk of fresh fruits, resulting in improved microbial safety as well as extending their shelf life. In general, it is recommended that, according to the effect of gamma radiation on physicochemical, microbial and sensorial characteristics, doses of 1.25-2 kGy could be used. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=antioxidant%20property" title="antioxidant property">antioxidant property</a>, <a href="https://publications.waset.org/abstracts/search?q=barberry%20fruit" title=" barberry fruit"> barberry fruit</a>, <a href="https://publications.waset.org/abstracts/search?q=chemical%20properties" title=" chemical properties"> chemical properties</a>, <a href="https://publications.waset.org/abstracts/search?q=gamma%20irradiation" title=" gamma irradiation"> gamma irradiation</a> </p> <a href="https://publications.waset.org/abstracts/81364/effects-of-gamma-irradiation-on-chemical-and-antioxidant-properties-of-iranian-native-fresh-barberry-fruit" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/81364.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">279</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2782</span> Studies on Induction of Cytotoxicity Through Apoptosis In Ovarian Cancer Cell Line (CAOV-3) by Chloroform Extract of Artocarpus Kemando Miq</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Noor%20Shafifiyaz%20Mohd%20Yazid">Noor Shafifiyaz Mohd Yazid</a>, <a href="https://publications.waset.org/abstracts/search?q=Najihah%20Mohd%20Hashim"> Najihah Mohd Hashim</a>, <a href="https://publications.waset.org/abstracts/search?q=Hapipah%20Mohd%20Ali"> Hapipah Mohd Ali</a>, <a href="https://publications.waset.org/abstracts/search?q=Syam%20Mohan"> Syam Mohan</a>, <a href="https://publications.waset.org/abstracts/search?q=Rosea%20Go"> Rosea Go</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Artocarpus kemando is a plant species from Moraceae family. This plant is used as household utensil by the local and the fruits are edible. The plants’ bark was used for the extraction process and yielded the chloroform crude extract which was used to screen for anticancer potential. The cytotoxic effect of the extract on CAOV-3 and WRL 68 cell lines were determined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide or MTT assays. Qualitative AO/PI assay was performed to confirm the apoptosis and necrosis process. Meanwhile, the measurement of cell loss, nuclear morphology, DNA content, cell membrane permeability, mitochondrial membrane potential changes and cytochrome c release from mitochondria were detected through cytotoxicity 3 assay. In MTT assay, A. kemando inhibited 50% growth of CAOV-3 cells at 27.9 ± 0:03, 20.1± 0:03, 18.21± 0:04 µg/mL after 24, 48 and 72 hour, respectively. The morphology changes can be seen on CAOV-3 with a production of cell membrane blebbing, cromatin condensation and apoptotic bodies. Evaluation of cytotoxicity 3 on CAOV-3 cells after treated with extract resulting loss of mitochondrial membrane potential and release of cytochrome c from mitochondria. The results demonstrated A. kemando has potentially anticancer agent, particularly on human ovarian cancer. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=anticancer" title="anticancer">anticancer</a>, <a href="https://publications.waset.org/abstracts/search?q=Artocarpus%20kemando" title=" Artocarpus kemando"> Artocarpus kemando</a>, <a href="https://publications.waset.org/abstracts/search?q=ovarian%20cancer" title=" ovarian cancer"> ovarian cancer</a>, <a href="https://publications.waset.org/abstracts/search?q=cytotoxicity" title=" cytotoxicity"> cytotoxicity</a> </p> <a href="https://publications.waset.org/abstracts/13699/studies-on-induction-of-cytotoxicity-through-apoptosis-in-ovarian-cancer-cell-line-caov-3-by-chloroform-extract-of-artocarpus-kemando-miq" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/13699.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">551</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2781</span> Design of a Remote Radiation Sensing Module Based on Portable Gamma Spectrometer</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Young%20Gil%20Kim">Young Gil Kim</a>, <a href="https://publications.waset.org/abstracts/search?q=Hye%20Min%20Park"> Hye Min Park</a>, <a href="https://publications.waset.org/abstracts/search?q=Chan%20Jong%20Park"> Chan Jong Park</a>, <a href="https://publications.waset.org/abstracts/search?q=Koan%20Sik%20Joo"> Koan Sik Joo</a> </p> <p class="card-text"><strong>Abstract:</strong></p> A personal gamma spectrometer has to be sensitive, pocket-sized, and carriable on the users. In our experimental study, we have explored the effectiveness of curcumin in protecting the structural paradigm of Human Serum Albumin (HSA) when exposed to gamma irradiation. HSA, being an important transport protein of the circulatory system, is involved in binding of variety of metabolites, drugs, dyes and fatty acids due to the presence of hydrophobic pockets inside the structure. HSA is also actively involved in the transportation of drugs and metabolites to their targets, because of its long half-life and regulation of osmotic blood pressure. Gamma rays, in its increasing concentration, results in structural alteration of the protein and superoxide radical generation. Curcumin, on the other hand, mitigates the damage, which has been evidenced in the following experiments. Our study explores the possibility for protection by curcumin during the molecular and conformational changes of HSA when exposed to gamma irradiation. We used a combination of spectroscopic methods to probe the conformational ensemble of the irradiated HSA and finally evaluated the extent of restoration by curcumin. SDS - PAGE indicated the formation of cross linked aggregates as a consequence of increasing exposure of gamma radiation. CD and FTIR spectroscopy inferred significant decrease in alpha helix content of HSA from 57% to 15% with increasing radiation doses. Steady state and time resolved fluorescence studies complemented the spectroscopic measurements when lifetime decay was significantly reduced from 6.35 ns to 0.37 ns. Hydrophobic and bityrosine study showed the effectiveness of curcumin for protection against radiation induced free radical generation. Moreover, bityrosine and hydrophobic profiling of gamma irradiated HSA in presence and absence of curcumin provided light on the formation of ROS species generation and the protective (magical) role of curcumin. The molecular mechanism of curcumin protection to HSA from gamma irradiation is yet unknown, though a possible explanation has been proposed in this work using Thioflavin T assay. It was elucidated, that when HSA is irradiated at low dose of gamma radiation in presence of curcumin, it is capable of retaining the native characteristic properties to a greater extent indicating stabilization of molecular structure. Thus, curcumin may be utilized as a therapeutic strategy to protect cellular proteins. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=Bityrosine%20content" title="Bityrosine content">Bityrosine content</a>, <a href="https://publications.waset.org/abstracts/search?q=conformational%20change" title=" conformational change"> conformational change</a>, <a href="https://publications.waset.org/abstracts/search?q=curcumin" title=" curcumin"> curcumin</a>, <a href="https://publications.waset.org/abstracts/search?q=gamma%20radiation" title=" gamma radiation"> gamma radiation</a>, <a href="https://publications.waset.org/abstracts/search?q=human%20serum%20albumin" title=" human serum albumin"> human serum albumin</a> </p> <a href="https://publications.waset.org/abstracts/77682/protective-role-of-curcumin-against-ionising-radiation-of-gamma-ray" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/77682.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">156</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">2778</span> Performance Analysis of M-Ary Pulse Position Modulation in Multihop Multiple Input Multiple Output-Free Space Optical System over Uncorrelated Gamma-Gamma Atmospheric Turbulence Channels</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Hechmi%20Saidi">Hechmi Saidi</a>, <a href="https://publications.waset.org/abstracts/search?q=Noureddine%20Hamdi"> Noureddine Hamdi</a> </p> <p class="card-text"><strong>Abstract:</strong></p> The performance of Decode and Forward (DF) multihop Free Space Optical ( FSO) scheme deploying Multiple Input Multiple Output (MIMO) configuration under Gamma-Gamma (GG) statistical distribution, that adopts M-ary Pulse Position Modulation (MPPM) coding, is investigated. There are some reports that IGRA results are affected by the patient's age. This study aims to compare the results of both IGRA tests according to age groups. Methods: We reviewed 54,882 samples referred to an independent reference laboratory (Seegene Medical Foundation, Seoul, Korea) for the diagnosis of LTBI from January 1, 2021, to December 31, 2021. This retrospective study enrolled 955 patients tested using QFT-GIT and 53,927 patients tested using T-SPOT.TB. The results of both IGRAs were divided in three age groups (0-9, 10-17, and ≥18-year old). The positive rates and the indeterminate rates between QFT-GIT and T-SPOT.TB were compared. We also evaluated the differences in positive and indeterminate rates by age-group. Results: The positive rate of QFT-GIT was 20.1% (192/955) and that of T-SPOT.TB was 8.7% (4704/53927) in overall patients. The positive rates of QFT-GIT in individuals aged 0-9, 10-17, and over 18-year old were 15.4%, 13.3%, and 22.0%, respectively. The positive rates of T-SPOT.TB were 8.9%, 2.0% and 8.8%,in each agegroup, respectively. The overall prevalence of indeterminate results was 2.1% (20/955) of QFT-GIT and 0.5% (270/53927) of T-SPOT.TB. The indeterminate rates of QFT-GIT in individuals aged 0-9, 10-17, and over 18 years were 0.4%, 6.7%, and 2.6%, respectively. The indeterminate rate of T-SPOT.TB were 0.5%, 0.7% and 0.5%,in each age group, respectively. Conclusion: Our findings suggest that T-SPOT.TB has a lower rate of positive results in overall patients and a lower rate of indeterminate results than those of QFT-GIT. The highest positive rate was found in the over 18 years group for QFT-GIT, but the positive rates of T-SPOT.TB was not significantly different among groups by age. QFT-GIT showed variable and higher indeterminate rates according to age group, but T-SPOT.TB showed lower rates in all age groups(<1%). <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=LTBI" title="LTBI">LTBI</a>, <a href="https://publications.waset.org/abstracts/search?q=IGRA" title=" IGRA"> IGRA</a>, <a href="https://publications.waset.org/abstracts/search?q=QFT-GIT" title=" QFT-GIT"> QFT-GIT</a>, <a href="https://publications.waset.org/abstracts/search?q=T-SPOT.%20TB" title=" T-SPOT.
