Past studies indicated that anemia, inflammatory process, and oxidative stress are interconnected. Erythrocytes are continuously exposed to reactive oxygen species (ROS) during circulation, due to normal aerobic cellular metabolism and also pathology of inflammatory diseases. Systemic mastocytosis and genetic depletion of mast cells have been shown to affect anaemia. In the present study, we attempted to reveal whether mast cells have a direct role in clearance or erythrophagocytosis of normal or oxidatively damaged erythrocytes. Murine erythrocytes were treated with tert-butyl hydroperoxidase (t-BHP), an agent that induces oxidative damage and mimics in vivo oxidative stress. Normal and oxidatively damaged erythrocytes were labeled with carboxyfluorescein succinimidyl ester (CFSE) to track erythrophagocytosis. We show, for the first time, direct erythrophagocytosis of oxidatively damaged erythrocytes in vitro by RBL-2H3 mast cells as well as in vivo by murine peritoneal mast cells. Also, activated mast cells, as may be present in inflammatory conditions, showed a significant increase in the uptake of oxidatively damaged erythrocytes than resting mast cells. This suggests the involvement of mast cells in erythrocyte clearance during oxidative stress or inflammatory disorders. Partial inhibition of phagocytosis by various inhibitors indicated that this process may be controlled by several pathways. Hence, our study provides important evidence for involvement of mast cells in severe anemia due to inflammation and oxidative stress and might be helpful to circumvent the adverse anemic disorders.
Keywords: mast cells, anemia, erythrophagocytosis, oxidatively damaged erythrocytes
Procedia PDF Downloads 254

487 Occult Haemolacria Paradigm in the Study of Tears

Authors: Yuliya Huseva

Abstract:
To investigate the contents of tears to determine latent blood. Methods: Tear samples from 72 women were studied with the microscopy of tears aspirated with a capillary and stained by Nocht and with a chemical method of test strips with chromogen. Statistical data processing was carried out using statistical packages Statistica 10.0 for Windows, calculation of Pearson's chi-square test, Yule association coefficient, the method of determining sensitivity and specificity. Results:, In 30.6% (22) of tear samples erythrocytes were revealed microscopically. Correlations between the presence of erythrocytes in the tear and the phase of the menstrual cycle has been discovered. In the follicular phase of the cycle, erythrocytes were found in 59.1% (13) people, which is significantly more (x2=4.2, p=0.041) compared to the luteal phase - in 40.9% (9) women. In the first seven days of the follicular phase of the menstrual cycle the erythrocytes were predominanted of in the tears of women examined testifies in favour of the vicarious bleeding from the mucous membranes of extragenital organs in sync with menstruation. Of the other cellular elements in tear samples with latent haemolacria, neutrophils prevailed - in 45.5% (10), while lymphocytes were less common - in 27.3% (6), because neutrophil exudation is accompanied by vasodilatation of the conjunctiva and the release of erythrocytes into the conjunctival cavity. It was found that the prognostic significance of the chemical method was 0.53 of the microscopic method. In contrast to microscopy, which detected blood in tear samples from 30.6% (22) of women, blood was detected chemically in tears of 16.7% (12). An association between latent haemolacria and endometriosis was found (k=0.75, p≤0.05). Microscopically, in the tears of patients with endometriosis, erythrocytes were detected in 70% of cases, while in healthy women without endometriosis - in 25% of cases. The proportion of women with erythrocytes in tears, determined by a chemical method, was 41.7% among patients with endometriosis, which is significantly more (x2=6.5, p=0.011) than 11.7% among women without endometriosis. The data obtained can be explained by the etiopathogenesis of the extragenital endometriosis which is caused by hematogenous spread of endometrial tissue into the orbit. In endometriosis, erythrocytes are found against the background of accumulations of epithelial cells. In the tear samples of 4 women with endometriosis, glandular cuboidal epithelial cells, morphologically similar to endometrial cells, were found, which may indicate a generalization of the disease. Conclusions: Single erythrocytes can normally be found in the tears, their number depends on the phase of the menstrual cycle, increasing in the follicular phase. Erythrocytes found in tears against the background of accumulations of epitheliocytes and their glandular atypia may indicate a manifestation of extragenital endometriosis. Both used methods (microscopic and chemical) are informative in revealing latent haemolacria. The microscopic method is more sensitive, reveals intact erythrocytes, and besides, it provides information about other cells. At the same time, the chemical method is faster and technically simpler, it determines the presence of haemoglobin and its metabolic products, and can be used as a screening.
Keywords: tear, blood, microscopy, epitheliocytes
Procedia PDF Downloads 120

486 Scanning Electron Microscopy of the Erythrocytes of Channa punctatus (Bloch) Exposed to Mercuric Chloride

Authors: Shweta Maheshwari, Anish Dua

Abstract:
Hematological changes reflect the adverse effects of heavy metals on fish. Hematology is a valuable tool to evaluate pathological condition of the fish. It helps in diagnosing the structural and functional status of fish exposed to toxicants. Morphological alteration in erythrocytes due to environmental stress can be studied through ultra-structural analysis. The aim of the present study was to assess the toxicity of mercuric chloride on red blood cells of an air breathing fish, Channa punctatus. Fish were subjected to chronic experiments using three sublethal concentration of mercuric chloride (0.020mg/L, 0.027mg/L, 0.040mg/L) for a period of 15, 30 and 60 days. Exposed fish of all the three concentrations were subjected to a recovery period of 30 days. A control was maintained in tap water simultaneously. For SEM analysis, blood from caudal vein of fish was taken and examined at an accelerating voltage of 20kV. Scanning electron micrographs revealed elliptical shaped erythrocytes of control fish. Alterations in the erythrocyte morphology such as presence of spherocytes, membrane internalization, crenation of membrane and development of lobopodial projections were observed in the exposed fish. The study revealed that ultra-structural analysis appears to be a sensitive method to evaluate the toxicity of various toxicants to fish.
Keywords: Channa punctatus, erythrocytes, mercuric chloride, scanning electron microscopy
Procedia PDF Downloads 371

485 Experimental Investigation of Damaged Reinforced Concrete Beams Repaired with Carbon Fibre Reinforced Polymer (CFRP) Strip under Impact Loading

Authors: M. Most of the damaged structures members such as beams, columns and slabs are not totally failed and it can be repaired. Nowadays, carbon fibre reinforced polymer CFRP has been wildly used in strengthening and retrofitting the structures members. CFRP can rector the load carrying capacity of the damaged structures members to make them serviceable. An experimental investigation was conducted to investigate the impact behaviour of the damaged beams repaired with CFRP. The tested beams had different degrees of damage and near surface mounted technique NSM was used to install the CFRP. A heavy drop weight impact test machine was used to conduct the experimental work. The study investigated the impact strength, stiffness, cracks and deflection of the CFRP repaired beams. The results show that CFRP significantly increased the impact resistance of the damaged beams. CFRP increased the damaged beams stiffness and reduced the deflection. The results showed that the NSM technique is more effective in repairing beams and preventing the debonding of the CFRP.
Keywords: damaged, concrete, impact, repaired
Procedia PDF Downloads 344

484 Analysis of Steel Beam-Column Joints Under Seismic Loads

Authors: Mizam Doğan

Abstract:
Adapazarı railway car factory, the only railway car factory of Turkey, was constructed in 1950. It was a steel design and it had filled beam sections and truss beam systems. Columns were steel profiles and box sections. The factory was damaged heavily on Izmit Earthquake and closed. In this earthquake 90% of damaged structures are reinforced concrete, the others are %7 prefabricated and 3% steel construction. As can be seen in statistical data, damaged industrial buildings in this earthquake were generally reinforced concrete and prefabricated structures. Adapazari railway car factory is the greatest steel structure damaged in the earthquake. This factory has 95% of the total damaged steel structure area. In this paper; earthquake damages on beams and columns of the factory are studied by considering TS648 'Turkish Standard Building Code for Steel Structures' and also damaged connection elements as welds, rivets and bolts are examined. A model similar to the damaged system is made and high-stress zones are searched. Free radicals from cigarette smoke can cause oxidative stress. The oxidative insults can lead to red blood cell (RBC) senescence and are involved in the clearance of red blood cells. The objective of the present study is to assess the association between smoke, oxidative stress evaluated with serum Malondialdehyde (MDA) level and phosphatidylserine (PS) externalization (biomarker of RBC senescence) evaluated with annexin V binding. A total of sixty-four male volunteers aged 25-60 years old were recruited in this study. MDA was measured by colorimetric method. Annexin V binding was detected by flow cytometry. Our results show that there was a significant increase in MDA levels in cigarette smokers as compared to non-smokers (p < 0.001). However, there was no significant different between annexin V binding (% gate) in cigarette smokers and non-smokers (p = 0.978). These results provide evidence of free radical from smoking is associated with oxidative damage to erythrocytes. However, our results suggest that PS externalization is unlikely to have a role in RBC senescence pathway of stressed erythrocytes from cigarette smoke. The other biomarker of RBC senescence should be determined on cigarette smoker erythrocytes.
Keywords: malondialdehyde, phosphatidylserine, RBC senescence, annexin V
Procedia PDF Downloads 437

482 The Effect of Bisphenol A and Its Selected Analogues on Antioxidant Enzymes Activity in Human Erythrocytes

Authors: Aneta Maćczak, Bożena Bukowska, Jaromir Michałowicz

Abstract:
Bisphenols are one of the most widely used chemical compounds worldwide. They are used in the manufacturing of polycarbonates, epoxy resins and thermal paper which are applied in plastic containers, bottles, cans, newspapers, receipt and other products. Among these compounds, bisphenol A (BPA) is produced in the highest amounts. There are concerns about endocrine impact of BPA and its other toxic effects including hepatotoxicity, neurotoxicity and carcinogenicity on human organism. Moreover, BPA is supposed to increase the incidence the obesity, diabetes and heart disease. For this reason the use of BPA in the production of plastic infant feeding bottles and some other consumers products has been restricted in the European Union and the United States. Nowadays, BPA analogues like bisphenol F (BPF) and bisphenol S (BPS) have been developed as alternative compounds. The replacement of BPA with other bisphenols contributed to the increase of the exposure of human population to these substances. Toxicological studies have mainly focused on BPA. In opposite, a small number of studies concerning toxic effects of BPA analogues have been realized, which makes impossible to state whether those substituents are safe for human health. Up to now, the mechanism of bisphenols action on the erythrocytes has not been elucidated. That is why, the aim of this study was to assess the effect of BPA and its selected analogues such as BPF and BPS on the activity of antioxidant enzymes, i.e. catalase (EC 1.11.1.6.), glutathione peroxidase (E.C.1.11.1.9) and superoxide dismutase (EC.1.15.1.1) in human erythrocytes. Red blood cells in respect to their function (transport of oxygen) and very well developed enzymatic and non-enzymatic antioxidative system, are useful cellular model to assess changes in redox balance. Erythrocytes were incubated with BPA, BPF and BPS in the concentration ranging from 0.5 to 100 µg/ml for 24 h. The activity of catalase was determined by the method of Aebi (1984). The activity of glutathione peroxidase was measured according to the method described by Rice-Evans et al. (1991), while the activity of superoxide dismutase (EC.1.15.1.1) was determined by the method of Misra and Fridovich (1972). The results showed that BPA and BPF caused changes in the antioxidative enzymes activities. BPA decreased the activity of examined enzymes in the concentration of 100 µg/ml. We also noted that BPF decreased the activity of catalase (5-100 µg/ml), glutathione peroxidase (50-100 µg/ml) and superoxide dismutase (25-100 µg/ml), while BPS did not cause statistically significant changes in investigated parameters. The obtained results suggest that BPA and BPF disrupt redox balance in human erythrocytes but the observed changes may occur in human organism only during occupational or subacute exposure to these substances.
Keywords: antioxidant enzymes, bisphenol A, bisphenol a analogues, human erythrocytes
Procedia PDF Downloads 471

481 The Effect of Total Mixture Concentrate Based on Tofu Waste Silage as Feed on Performance of Lambs

Authors: Yafri Hazbi, Zaenal Bachruddin, Nafiatul Umami, Lies Mira Yusiati

Abstract:
The objective of this study was to identify the benefits of total mixture concentrate based on tofu waste silage (TMC-TWS) as ration containing lactic acid bacteria on performance of lambs. Fifteen weaning lambs (2-3 months old) were randomly divided into two treatment groups, treatment group I (TI) was fed with TMC-TWS as ration and treatment group II (TII) was fed with TMC-TWS fresh (without silage fermentation). The performance of lambs was evaluated on day 0, 15, and 30 to have data of body weight per day. Meanwhile, blood sampling and feces were made on the 30th day to get an analysis on the blood profile (erythrocytes (mill/ml), hemoglobin (g/dL), packed cell volume (%), and leukocytes (mill/ml)) and the number of worm eggs in feces. The results of this study showed no significant difference between the effect of different feed on the blood profile (erythrocytes (mill/ml), hemoglobin (g/dL), packed cell volume (%), as well as the number of worm eggs in the feces. However the results showed significant differences if it is low (P<0.05) due to the treatment group based on sex on body weight gain per day, feed conversion rate and the number of erythrocytes.
Keywords: lambs, total mixture concentrate, silage, acid lactid bacteria, blood profile, eggs worm in feces
Procedia PDF Downloads 178

480 Investigation of Rehabilitation Effects on Fire Damaged High Strength Concrete Beams

Authors: Eun Mi Ryu, Ah Young An, Ji Yeon Kang, Yeong Soo Shin, Hee Sun Kim

Abstract:
As the number of fire incidents has been increased, fire incidents significantly damage economy and human lives. Especially when high strength reinforced concrete is exposed to high temperature due to a fire, deterioration occurs such as loss in strength and elastic modulus, cracking, and spalling of the concrete. Therefore, it is important to understand risk of structural safety in building structures by studying structural behaviors and rehabilitation of fire damaged high strength concrete structures. This paper aims at investigating rehabilitation effect on fire damaged high strength concrete beams using experimental and analytical methods. In the experiments, flexural specimens with high strength concrete are exposed to high temperatures according to ISO 834 standard time temperature curve. After heated, the fire damaged reinforced concrete (RC) beams having different cover thicknesses and fire exposure time periods are rehabilitated by removing damaged part of cover thickness and filling polymeric mortar into the removed part. From four-point loading test, results show that maximum loads of the rehabilitated RC beams are 1.8~20.9% higher than those of the non-fire damaged RC beam. On the other hand, ductility ratios of the rehabilitated RC beams are decreased than that of the non-fire damaged RC beam. In addition, structural analyses are performed using ABAQUS 6.10-3 with same conditions as experiments to provide accurate predictions on structural and mechanical behaviors of rehabilitated RC beams. For the rehabilitated RC beam models, integrated temperature–structural analyses are performed in advance to obtain geometries of the fire damaged RC beams. After spalled and damaged parts are removed, rehabilitated part is added to the damaged model with material properties of polymeric mortar. Three dimensional continuum brick elements are used for both temperature and structural analyses. The same loading and boundary conditions as experiments are implemented to the rehabilitated beam models and nonlinear geometrical analyses are performed. Structural analytical results show good rehabilitation effects, when the result predicted from the rehabilitated models are compared to structural behaviors of the non-damaged RC beams. In this study, fire damaged high strength concrete beams are rehabilitated using polymeric mortar. From four point loading tests, it is found that such rehabilitation is able to make the structural performance of fire damaged beams similar to non-damaged RC beams. The predictions from the finite element models show good agreements with the experimental results and the modeling approaches can be used to investigate applicability of various rehabilitation methods for further study.
Keywords: fire, high strength concrete, rehabilitation, reinforced concrete beam
Procedia PDF Downloads 445

479 Forced Swim Stress Does Not Induce Structural Chromosomal Aberrations in Rat Bone Marrow

Authors: Mohammad Y. Moreover, this research is focused on the influence of hyaluronic acid (HA) on UVA treated dermal fibroblasts. We aim to establish whether HA can effectively relief ECM degradation, and restore the elasticity of UVA-damaged fibroblasts. Prolonged exposure to UVA radiation can damage fibroblasts and led variation in cell morphology and reduction in cell viability. Besides, UVA radiation can induce IL-1β expression on fibroblasts and then promote MMP-1 and MMP-3 expression, which can accelerate ECM degradation. On the other hand, prolonged exposure to UVA radiation reduced collagen and elastin synthesis on fibroblasts. Due to the acceleration of ECM degradation and the reduction of ECM synthesis, Atomic force microscope (AFM) was used to analyze the elasticity reduction on UVA-damaged fibroblasts. UVA irradiation causes photoaging on fibroblasts. UVA damaged fibroblasts with HA treatment can down-regulate the gene expression of MMP-1, MMP-3, and then slow down ECM degradation. On the other hand, HA may restore elastin and collagen synthesis in UV-damaged fibroblasts. Based on the slowdown of ECM degradation, UVA-damaged fibroblast elasticity can be effectively restored by HA treatment. In summary, HA can relief the photoaging conditions on fibroblasts, but may not be able to return fibroblasts to normal, healthy state. Although HA cannot fully recover UVA-damaged fibroblasts, HA is still potential for repairing photoaging skin.
Keywords: atomic force microscope, hyaluronic acid, UVA radiation, dermal fibroblasts
Procedia PDF Downloads 391

477 Deformation Characteristics of Fire Damaged and Rehabilitated Normal Strength Concrete Beams

Authors: Yeo Kyeong Lee, Hae Won Min, Ji Yeon Kang, Hee Sun Kim, Yeong Soo Shin

Abstract:
Fire incidents have been steadily increased over the last year according to national emergency management agency of South Korea. Even though most of the fire incidents with property damage have been occurred in building, rehabilitation has not been properly done with consideration of structure safety. Therefore, this study aims at evaluating rehabilitation effects on fire damaged normal strength concrete beams through experiments and finite element analyses. For the experiments, reinforced concrete beams were fabricated having designed concrete strength of 21 MPa. Two different cover thicknesses were used as 40 mm and 50 mm. After cured, the fabricated beams were heated for 1hour or 2hours according to ISO-834 standard time-temperature curve. Rehabilitation was done by removing the damaged part of cover thickness and filling polymeric mortar into the removed part. Both fire damaged beams and rehabilitated beams were tested with four point loading system to observe structural behaviors and the rehabilitation effect. To verify the experiment, finite element (FE) models for structural analysis were generated using commercial software ABAQUS 6.10-3. For the rehabilitated beam models, integrated temperature-structural analyses were performed in advance to obtain geometries of the fire damaged beams. In addition to the fire damaged beam models, rehabilitated part was added with material properties of polymeric mortar. Three dimensional continuum brick elements were used for both temperature and structural analyses. The same loading and boundary conditions as experiments were implemented to the rehabilitated beam models and non-linear geometrical analyses were performed. Test results showed that maximum loads of the rehabilitated beams were 8~10% higher than those of the non-rehabilitated beams and even 1~6 % higher than those of the non-fire damaged beam. Stiffness of the rehabilitated beams were also larger than that of non-rehabilitated beams but smaller than that of the non-fire damaged beams. In addition, predicted structural behaviors from the analyses also showed good rehabilitation effect and the predicted load-deflection curves were similar to the experimental results. From this study, both experiments and analytical results demonstrated good rehabilitation effect on the fire damaged normal strength concrete beams. For the further, the proposed analytical method can be used to predict structural behaviors of rehabilitated and fire damaged concrete beams accurately without suffering from time and cost consuming experimental process.
Keywords: fire, normal strength concrete, rehabilitation, reinforced concrete beam
Procedia PDF Downloads 508

476 Investigation of the Effects of Aerobic Exercise Programs on Hematological Parameters of Sedentary People

Authors: Sanjeev Kumar, Swati Choudhary

Abstract:
Background: A variety of studies warn that sedentary lifestyles can contribute to many preventable causes of death. This study was taken to determine the effects of two types of aerobic training programs on erythrocytes, leukocytes, hemoglobin concentration (Hb), platelets and hematocrit of sedentary people (N=60) with age group 20 to 30 years. Methods: All the subjects were randomly divided into three groups i.e. two experiments groups (aerobic dance & cardio fitness) and control group. Each group having 10 male and 10 females. Experimental groups undergone 60 minutes of training 5 times a week for 12 weeks whereas the control group did not participate in any training program except their daily routine. The aerobic dance group was chosen to perform exercise like step –touch, side-to-side, V-step and hand and body movements, etc. The cardio fitness group was chosen to perform exercises with modern fitness equipment like treadmill, elliptical trainer, stationary bike and rowing machine. Rating of perceived exertion (RPE) scale developed by Gunner Borg was used to monitor the intensity of the workout. Aerobic programs were encompassed of low-impact (0- 4 week & perceived exertion from 6 to 12), moderate-impact (4-8 week and perceived exertion from 12 to 16) and high-impact (8- 12 week & perceived exertion from 16 to 20). Results: To test the effectiveness of training programs paired t-test was used and significant difference (p<0.05) was observed in erythrocytes, hemoglobin concentration, platelets, hematocrit but no significant effects of training was found in leukocytes (p>0.05). Paired t-test also showed that no effect of time was seen in the control group in all the cases (p>0.05). Further analysis of covariance was used to know which program was more effective and it was seen that F value was found significant in the case of erythrocytes, hemoglobin concentration, platelets, and hematocrit as their associated p-value (p<0.05) is lesser than 0.05. As F value was found significant for hematological parameters, fishers least significant difference test was used and results of post hoc mean comparison indicated that experimental groups (aerobic dance group and cardio fitness group) had significant difference with control group in erythrocytes, hemoglobin concentration, platelets and hematocrit and insignificant difference was found between aerobic dance group & cardio fitness group in all the cases. Thus, it may be concluded that in general, both the aerobic training programs had adequate effects on all the hematological parameters except leukocytes.
Keywords: aerobic dance, cardio fitness, hematological variables, rating perceived exertion scale
Procedia PDF Downloads 272

475 Numerical and Comparative Analysis between Two Composite Plates Notched in Different Shapes and Repaired by Composite

Authors: Amari Khaoula, Berrahou Mohamed

Abstract:
The topic of our article revolves around a numerical and comparative analysis between two notched Boron/epoxy plates that are U-shaped and the other V-shaped, cracked, and repaired by a rectangular patch of the same composite material; the finite element method was used for the analytical study and comparison of the results obtained for determining the optimal shape of notch which will give a longer life to the repair. Finally, three actual earthquake-damaged school buildings in Taiwan are used as a case study to demonstrate application of the proposed assessment method.
Keywords: seismic assessment, seismic reduction factor, residual seismic ratio, post-earthquake, reinforced concrete, building
Procedia PDF Downloads 400

473 Post-Earthquake Road Damage Detection by SVM Classification from Quickbird Satellite Images

Authors: Moein Izadi, Ali Mohammadzadeh

Abstract:
Detection of damaged parts of roads after earthquake is essential for coordinating rescuers. In this study, an approach is presented for the semi-automatic detection of damaged roads in a city using pre-event vector maps and both pre- and post-earthquake QuickBird satellite images. Damage is defined in this study as the debris of damaged buildings adjacent to the roads. Some spectral and texture features are considered for SVM classification step to detect damages. Finally, the proposed method is tested on QuickBird pan-sharpened images from the Bam City earthquake and the results show that an overall accuracy of 81% and a kappa coefficient of 0.71 are achieved for the damage detection. The obtained results indicate the efficiency and accuracy of the proposed approach.
Keywords: SVM classifier, disaster management, road damage detection, quickBird images
Procedia PDF Downloads 623

472 Red Blood Cells Deformability: A Chaotic Process

Authors: Ana M. An electro-optic mechanic system called erythrodeformeter has been developed and constructed in our laboratory in order to evaluate the erythrocytes' viscoelasticity. A numerical method formulated on the basis of fractal approximation for ordinary (OBM) and fractionary Brownian motion (FBM), as well as wavelet transform analysis, are proposed to distinguish chaos from noise based on the assumption that diffractometric data involves both deterministic and stochastic components, so it could be modelled as a system of bounded correlated random walk. Here we report studies on 25 donors: 4 alpha thalassaemic patients, 11 beta thalassaemic patients, and 10 healthy controls non-alcoholic and non-smoker individuals. The Correlation Coefficient, a nonlinear parameter, showed evidence of the changes in the erythrocyte deformability; the Wavelet Entropy could quantify those differences which are detected by the light diffraction patterns. Such quantifiers allow a good deal of promise and the possibility of a better understanding of the rheological erythrocytes aspects and also could help in clinical diagnosis.
Keywords: red blood cells, deformability, nonlinear dynamics, chaos theory, wavelet trannsform
Procedia PDF Downloads 59

471 Genotoxic and Cytotoxic Effects of Methidathion Pesticide

Authors: Mohammad Y. CA, MI (number of cells undergoing mitosis) necrotic, and apoptotic cells recorded in lymphocytes. Results showed that there was a slight increase in the frequency of micronucleated bone marrow cells. However, no structural chromosomal aberrations were detected in vivo or in vitro. On the other hand, the results showed significant increase in necrotic and apoptotic cells following MTD administration in a dose-dependent manner comparing to positive and negative control groups. In light of these results, MTD can be considered highly cytotoxic and moderate genotoxic, and precaution should be taken when using MTD.
Keywords: methidathion, micronucleus, NDI, NDCI, toxicity, chromosomal aberrations
Procedia PDF Downloads 412

470 Assessment of the Number of Damaged Buildings from a Flood Event Using Remote Sensing Technique

Authors: Jaturong Som-ard

Abstract:
The heavy rainfall from 3rd to 22th January 2017 had swamped much area of Ranot district in southern Thailand. Due to heavy rainfall, the district was flooded which had a lot of effects on economy and social loss. The major objective of this study is to detect flooding extent using Sentinel-1A data and identify a number of damaged buildings over there. The data were collected in two stages as pre-flooding and during flood event. Calibration, speckle filtering, geometric correction, and histogram thresholding were performed with the data, based on intensity spectral values to classify thematic maps. The maps were used to identify flooding extent using change detection, along with the buildings digitized and collected on JOSM desktop. The numbers of damaged buildings were counted within the flooding extent with respect to building data. The total flooded areas were observed as 181.45 sq.km. These areas were mostly occurred at Ban khao, Ranot, Takhria, and Phang Yang sub-districts, respectively. The Ban khao sub-district had more occurrence than the others because this area is located at lower altitude and close to Thale Noi and Thale Luang lakes than others. The numbers of damaged buildings were high in Khlong Daen (726 features), Tha Bon (645 features), and Ranot sub-district (604 features), respectively. The final flood extent map might be very useful for the plan, prevention and management of flood occurrence area. The map of building damage can be used for the quick response, recovery and mitigation to the affected areas for different concern organization.
Keywords: flooding extent, Sentinel-1A data, JOSM desktop, damaged buildings
Procedia PDF Downloads 192

469 Urban Search, Rescue and Rapid Field Assessment of Damaged and Collapsed Building Structures

Authors: Abid I. The nature of the work undertaken by USAR means that staying out of a damaged or collapsed building structure is not usually an option for search and rescue personnel. As a result, there is always a risk that they could become victims. For this paper, a systematic and investigative review using desk research was undertaken to explore the role which structural engineering can play in assisting search and rescue personnel to conduct structural assessments when in the field. The focus is on how search and rescue personnel can assess damaged and collapsed building structures, not just in terms of the structural damage that may be countered, but also in relation to structural stability. Natural disasters, accidental emergencies, acts of terrorism and other extreme events can vary significantly in nature and ferocity, and can cause a wide variety of damage to building structures. It is not possible or, even realistic, to provide search and rescue personnel with definitive guidelines and procedures to assess damaged and collapsed building structures as there are too many variables to consider. However, understanding what implications damage may have upon the structural stability of a building structure will enable search and rescue personnel to judge better and quantify the risk from a life-safety standpoint. It is intended that this will allow search and rescue personnel to make informed decisions and ensure every effort is made to mitigate risk so that they do not become victims.
Keywords: damaged and collapsed building structures, life safety, quantifying risk, search and rescue personnel, structural assessments in the field
Procedia PDF Downloads 392

468 Sustainability: Effect of Earthquake in Micro Hydro Sector, a Case Study of Micro Hydro Projects in Northern Part of Kavre District, Nepal

Authors: Ram Bikram Thapa, Ganesh Lama

Abstract:
The Micro Hydro is one of the successful technology in Rural Nepal. Four hundred and twenty fish samples were collected from six sites along the whole course of the river Nile, Egypt. The mean values of erythrocytes, thrombocytes, hemoglobin concentration, hematocrit value, and mean corpuscular volume were significantly lower in the blood of Nile tilapia and African catfish collected from downstream (contaminated) compared to upstream sites. In contrast, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration in the peripheral blood of both fish species significantly increased from upstream to downstream river Nile. The leukocytes count was significantly decreased in contaminated sites compared to upstream area. Hematological variables in the peripheral blood of Oreochromis niloticus niloticus and Clarias gariepinus exhibited significant (p<0.05) correlation with nearly all the detected chemical and physical parameters along the Nile course. In the present study, lower cellular and nuclear areas and cellular and nuclear shape factor were recorded in the erythrocytes of fish collected from downstream compared to those caught from upstream sites. This was confirmed by higher immature ratios of red cells in the blood of fish sampled from downstream river Nile. Karyorrhetic and enucleated erythrocytes were significantly correlated with physiochemical parameters in water samples collected from the same sites is being higher in the blood of fish collected from downstream sites. To see if there was any correlation between fish altered physiological fitness and environmental stress, we measured serum biochemical variables namely; total protein, cholesterol, triglycerides, calcium, chlorides, alkaline phosphatase activity (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), uric acid activity, creatinine, and serum glucose. The level of all the selected biochemical variables in the blood of O. niloticus niloticus and C. gariepinus were recorded to be significantly higher (p<0.05) in downstream sites. According to the present results, nearly all the detected haematological and blood biochemical variables are suitable indicators of contaminant exposure in O. niloticus niloticus and C. gariepinus. Also the detected erythrocytes malformations in blood collected from Nile tilapia and African catfish were proven to be suitable for bio-monitoring aquatic pollution. The results revealed species-specific differences in sensitivities, suggesting that Nile tilapia may serve as a more sensitive test species compared to African catfish.
Keywords: biomarkers, water pollution, blood parameters, river nile, african catfish, nile tilapia
Procedia PDF Downloads 291

465 Reconstructing Calvarial Bone Lesions Using PHBV Scaffolds and Cord Blood Mesenchymal Stem Cells in Rat

Authors: Hamed Hosseinkazemi, Esmaeil Biazar

Abstract:
For tissue engineering of bone, anatomical and operational reconstructions of damaged tissue seem to be vital. In this study the condition of biodegradable bed Nanofibrous PHBV and USSC cells were used to accelerate bone repair of damaged area. Hollow nanofabrication scaffold of damageable life was designed as PHBV by electrospinning and via determining the best factors such as the kind and amount of solvent, specific volume and rate. The separation of osseous tissue infiltration and evaluating its nature by flow cytometrocical analysis was done. Animal test including USSC as well as PHBV condition in the damaged bone was done in the rat. After 8 weeks the implanted area was analyzed using CT scan and was sent to histopathology ward. Finally, the rate and quality of reconstruction were determined after H and E coloring. Histomorphic analysis indicated a statistically significant difference between the experimental group of PHBV, USSC+PHBV and control group. Besides, the histopathologic analysis showed that bone reconstruction rate was high in the area containing USSC and PHBV, compared with area having PHBV and control group and consequently the reconstruction quality of bones and the relationship between the new bone tissues and surrounding bone tissues were high too. Using PHBR scaffold and USSC together could be useful in the amending of wide range of bone lesion.
Keywords: bone lesion, nanofibrous PHBV, stem cells, umbilical cord blood
Procedia PDF Downloads 317

464 Overview and Post Damage Analysis of Nepal Earthquake 2015

Authors: Vipin Kumar Singhal, Rohit Kumar Mittal, Pavitra Ranjan Maiti

Abstract:
Damage analysis is one of the preliminary activities to be done after an earthquake so as to enhance the seismic building design technologies and prevent similar type of failure in future during earthquakes. This research article investigates the damage pattern and most probable reason of failure by observing photographs of seven major buildings collapsed/damaged which were evenly spread over the region during Mw7.8, Nepal earthquake 2015 followed by more than 400 aftershocks of Mw4 with one aftershock reaching a magnitude of Mw7.3. Over 250,000 buildings got damaged, and more than 9000 people got injured in this earthquake. Photographs of these buildings were collected after the earthquake and the cause of failure was estimated along with the severity of damage and comment on the reparability of structure has been made. Based on observations, it was concluded that the damage in reinforced concrete buildings was less compared to masonry structures. The number of buildings damaged was high near Kathmandu region due to high building density in that region. This type of damage analysis can be used as a cost effective and quick method for damage assessment during earthquakes.
Keywords: Nepal earthquake, damage analysis, damage assessment, damage scales
Procedia PDF Downloads 374

463 Experimental and Analytical Studies for the Effect of Thickness and Axial Load on Load-Bearing Capacity of Fire-Damaged Concrete Walls

Authors: Yeo Kyeong Lee, Ji Yeon Kang, Eun Mi Ryu, Hee Sun Kim, Yeong Soo Shin

Abstract:
The objective of this paper is an investigation of the effects of the thickness and axial loading during a fire test on the load-bearing capacity of a fire-damaged normal-strength concrete wall. Two factors are attributed to the temperature distributions in the concrete members and are mainly obtained through numerous experiments. Toward this goal, three wall specimens of different thicknesses are heated for 2 h according to the ISO-standard heating curve, and the temperature distributions through the thicknesses are measured using thermocouples. In addition, two wall specimens are heated for 2 h while simultaneously being subjected to a constant axial loading at their top sections. The test results show that the temperature distribution during the fire test depends on wall thickness and axial load during the fire test. After the fire tests, the specimens are cured for one month, followed by the loading testing. The heated specimens are compared with three unheated specimens to investigate the residual load-bearing capacities. The fire-damaged walls show a minor difference of the load-bearing capacity regarding the axial loading, whereas a significant difference became evident regarding the wall thickness. To validate the experiment results, finite element models are generated for which the material properties that are obtained for the experiment are subject to elevated temperatures, and the analytical results show sound agreements with the experiment results. The analytical method based on validated thought experimental results is applied to generate the fire-damaged walls with 2,800 mm high considering the buckling effect: typical story height of residual buildings in Korea. The models for structural analyses generated to deformation shape after thermal analysis. The load-bearing capacity of the fire-damaged walls with pin supports at both ends does not significantly depend on the wall thickness, the reason for it is restraint of pinned ends. The difference of the load-bearing capacity of fire-damaged walls as axial load during the fire is within approximately 5 %.
Keywords: normal-strength concrete wall, wall thickness, axial-load ratio, slenderness ratio, fire test, residual strength, finite element analysis
Procedia PDF Downloads 215

462 Flexural Behavior of Heat-Damaged Concrete Beams Reinforced with Fiber Reinforced Polymer (FRP) Bars

Authors: Mohammad R. In the past decades, fiber reinforced polymer (FRP) bars had been widely used to substitute the steel bars due to their high resistance to corrosion, high tensile capacity, and low weight in comparison with steel. Experimental studies on the behavior of FRP bar reinforced concrete beams had been carried out worldwide for a few decades. While the research on such structural members under elevated temperatures is still very limited. In this research, the flexural behavior of heat-damaged concrete beams reinforced with FRP bars is studied. Two types of FRP rebar namely, carbon fiber reinforced polymer (CFRP) and glass fiber reinforced polymer (GFRP), are used. The beams are subjected to four levels of temperature before tested to monitor their flexural behavior. The results are compared with other concrete beams reinforced with regular steel bars. The results show that the beams reinforced with CFRP bars and GFRP bars had higher flexural capacity than the beams reinforced with steel bars even if heated up to 400°C and 300°C, respectively. After that the beams reinforced with steel bars had the superiority.
Keywords: concrete beams, FRP rebar, flexural behavior, heat-damaged
Procedia PDF Downloads 443

461 Cellular Components of the Hemal Node of Egyptian Cattle

Authors: Amira E. Light microscopy revealed a capsule of connective tissue consisted mainly of collagen fiber surrounding hemal node, numerous erythrocytes were found in wide subcapsular sinus under the capsule. The parenchyma of the hemal node was divided into cortex and medulla. Diffused lymphocytes, and lymphoid follicles, having germinal centers were the main components of the cortex, while in the medulla there was wide medullary sinus, diffused lymphocytes and few lymphoid nodules. The area occupied with lymph nodules was larger than that occupied with non-nodular structure of lymphoid cords and blood sinusoids. Electron microscopy revealed the cellular components of hemal node including elements of circulating erythrocytes intermingled with lymphocytes, plasma cells, mast cells, reticular cells, macrophages, megakaryocytes and endothelial cells lining the blood sinuses. The lymphocytes were somewhat triangular in shape with cytoplasmic processes extending between adjacent erythrocytes. Nuclei were triangular to oval in shape, lightly stained with clear nuclear membrane indentation and clear nucleoli. The reticular cells were elongated in shape with cytoplasmic processes extending between adjacent lymphocytes, rough endoplasmic reticulum, ribosomes and few lysosomes were seen in their cytoplasm. Nucleus was elongated in shape with less condensed chromatin. Plasma cells were oval to irregular in shape with numerous dilated rough endoplasmic reticulum containing electron lucent material occupying the whole cytoplasm and few mitochondria were found. Nuclei were centrally located and oval in shape with heterochromatin emarginated and often clumped near the nuclear membrane. Occasionally megakaryocytes and mast cells were seen among lymphocytes. Megakaryocytes had multilobulated nucleus and free ribosomes often appearing as small aggregates in their cytoplasm, while mast cell had their characteristic electron dense granule in the cytoplasm, few electron lucent granules were found also, we conclude that, the main function of the hemal node of cattle is proliferation of lymphocytes. No role for plasma cell in erythrophagocytosis could be suggested.
Keywords: cattle, electron microscopy, hemal node, histology, immune system
Procedia PDF Downloads 402

460 Implementation of Tissue Engineering Technique to Nursing of Unhealed Diabetic Foot Lesion

Authors: Basuki Supartono

Abstract:
Introduction: Diabetic wound risks limb amputation, and the healing remains challenging. In sickle cell patients, adhesion of erythrocytes or reticulocytes to one another and the vessel wall results in painful ischemic episodes with few, if any, effective treatments for vaso-occlusive crises. Identification of disease-associated adhesion markers on erythrocytes or reticulocytes might inform the use of more effective therapies against vaso-occlusive crises. Increased expression of one or more of bcam, itga4, cd44, cd47, rap1a, vcam1, or icam4 has been reported in sickle cell subjects. Using the miRNet ontology knowledgebase, peripheral blood interactomes were generated by seeding various combinations of the afore-referenced mRNA. These interactomes yielded an array of miR targets. As examples, targeting hsa-miR-155-5p can potentially neutralize the rap1a-bcam-cd44-itga4-vcam1 erythrocyte/reticulocyte adhesion interactome whereas targeting hsa-miRs-103a-3p or 107 can potentially neutralize adhesion in cells overexpressing icam4-cd47-bcam-itga4-cd36. AM3380 (MIRacle™) is an off-the shelf hsa-miR-155-5p agomiR that can potentially neutralize the rap1a-bcam-cd44-itga4-vcam1 signaling axis. Phlebotomy coupled with transcriptomics represents a potentially feasible and effective precision medicine strategy to mitigate vaso-occlusive crises in sickle cell patients. <p
