The overall loss of total tocopherols during these stages was found to be 37.9%, although the general reduction trend of delta (d), gamma (c), and alpha (a) tocopherols is very similar during neutralization (35.3%), bleaching (38.3%), and deodorization (37.8%). However, in contrast to the neutralizing and deodorizing stages, the bleaching process caused relatively less reduction for individual tocopherol contents. Deodorizer distillates were also analyzed and were found to be rich with tocopherols content (29,348.24 lg/ml). The results of the study indicated that most parts of the tocopherols are wasted during processing. Therefore, the proper concentration of nutritionists, industrialists, and manufacturers is needed for the necessary improvements in processing technology to avoid the major loss of tocopherols and to mature sunflower through wet grinding followed by filtration then centrifugation and recovered as the buoyant fraction. Washing this fraction with buffer (water-washed oil bodies, WWOB) or 9 M urea (urea-washed oil bodies, UWOB) resulted in the removal of extraneous proteins. SDS-PAGE of the proteins still associated with the oil body fraction after washing indicated that this effect was particularly dramatic with urea washing. Thirty-eight percent of the total seed tocopherol was recovered in WWOB after only one cycle of oil body recovery. The total phenolic content (TPC) of differentially washed sunflower seed oil bodies was used as a marker for the nonspecific association of phenolic compounds to oil bodies. This value decreased with increased removal of proteins from oil bodies, whereas the converse was true for tocopherol values, which increased from 214 mg total tocopherol kg−1 WWOB [dry wt basis (dwb)] to 392 mg total tocopherol kg−1 UWOB (dwb). The ratio of the four tocopherol isomers remained constant in the seed and oil body preparations (α:β:γ:δ approximately 94∶5∶0.5∶0.5). Herein, rapid, sensitive and precise method for the determination of all type of tocopherols in different vegetable oils includinhg sunflower oil, olive oil and extra virgin olive oil is reported. Reverse Phase High Performance Liquid Chromatography (RP-HPLC) with UV detector was used for tocopherol determination. A simple, quick and sensitive method to estimate the antioxidant quantity is proposed. In this method oils were diluted in methanol and injected directly into column (no saponification procedure). Methanol and water (98:2) mixture was used as mobile phase. Three tocopherols (α, γ and δ) were detected at 292 nm wavelength with UV detector. Method had good limit of detection (LOD) (7ng/g) and reproducibility (C.V% 0.9, 0.8 and 0.4 for α, γ and δ tocopherols, respectively). Result showed the best source for α-tocopherol was sun flower oil (146.65±1.7mg/kg). Oil richest in δ-tocopherol was olive oil and δ-tocopherol was absent in extra virgin olive oil.
