During my PhD programme, my research contributions focused on molecular cell biology, specifically delving into the expression patterns of leptin (an Ob gene product) and the cytokines tumour necrosis factor-alpha and interleukins 6 and 8 in the reproductive system, with a particular emphasis on polycystic ovarian syndrome within the Indian population. With 18 years of hands-on experience as a molecular biologist, I have honed my expertise in the innovative CRISPR/Cas9 technology for genome editing. Notably, my proficiency extends to designing targeting and validation strategies for creating mutant mice and other organisms. I have successfully executed gene knockouts, introduced stop codons, implemented N-terminus GFP fusions, and incorporated short mutations like SNPs or HA tags. Over the span of 8 years, I have demonstrated my capabilities by producing three modified mice for experiments and validated projects. Beyond genome editing, I possess a comprehensive understanding of next-generation sequencing (NGS), with a particular focus on the IonTorrent sequencing platform. My knowledge encompasses all aspects, from sample quality control and library construction to operating instruments, assessing runs, and interpreting results. Additionally, I am well-versed in Pac Bio and Illumina platforms, offering consultation on experimental design, library preparation, and result interpretation. Proficient in various software tools such as Benchling (for Sanger sequencing analysis, CRISPR targeting designs, PCR, and qPCR primer design), Ion Reporter for Ion Torrent sequencing, Galaxy, R/R Studio, TrinityRNAseq, and Limma for RNAseq analysis, I bring a wealth of technological expertise to the table.
