Glabra (Kochummen), was taken under phytochemical investigation. The crude methanolic extract was partitioned with a different solvent system by increasing their polarities (n-hexane, dichloromethane, and methanol). The compounds were fractionated and isolated from n-hexane partition by using column chromatography with silica gel 60 or Sephadex LH-20 as a stationary phase and preparative thin layer chromatographic technique. Isolates were characterized using TLC, FTIR, UV spectrophotometer and NMR spectroscopy. The n-hexane fractionates yielded a total of four compounds namely N-methyllaurotetanine (1), dicentrine (2), β-sitosterol (3), and stigmasterol (4). The result indicates that the leaves of Actinodaphne sesquipedalis may provide a rich source of alkaloids and triterpenoids. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=actinodaphne%20sesquipedalis" title="actinodaphne sesquipedalis">actinodaphne sesquipedalis</a>, <a href="https://publications.waset.org/abstracts/search?q=alkaloids" title=" alkaloids"> alkaloids</a>, <a href="https://publications.waset.org/abstracts/search?q=phytochemical%20investigation" title=" phytochemical investigation"> phytochemical investigation</a>, <a href="https://publications.waset.org/abstracts/search?q=triterpenoids" title=" triterpenoids"> triterpenoids</a> </p> <a href="https://publications.waset.org/abstracts/20389/isolation-and-identification-of-compounds-from-the-leaves-of-actinodaphne-sesquipedalis-hook-f-var-glabra-lauraceae" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/20389.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">397</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">12</span> Cholinesterase Inhibitory Indole Alkaloids from the Bark of Rauvolfia reflexa</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Mehran%20Fadaeinasab">Mehran Fadaeinasab</a>, <a href="https://publications.waset.org/abstracts/search?q=Alireza%20Basiri"> Alireza Basiri</a>, <a href="https://publications.waset.org/abstracts/search?q=Yalda%20Kia"> Yalda Kia</a>, <a href="https://publications.waset.org/abstracts/search?q=Hamed%20Karimian"> Hamed Karimian</a>, <a href="https://publications.waset.org/abstracts/search?q=Hapipah%20Mohd%20Ali"> Hapipah Mohd Ali</a>, <a href="https://publications.waset.org/abstracts/search?q=Vikneswaran%20Murugaiyah"> Vikneswaran Murugaiyah</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Two new, rauvolfine C and 3- methyl-10,11-dimethoxyl-6- methoxycarbonyl- β- carboline, along with five known indole alkaloids, macusine B, vinorine, undulifoline, isoresrpiline and rescinnamine were isolated from the bark of Rauvolfia reflexa. All the compounds showed good to moderate cholinesterase inhibitory activity with IC50 values in the range of 8.06 to 73.23 πM, except rauvolfine C that was inactive against acetylcholinesterase (AChE). Rescinnamine, a dual inhibitor was found to be the most potent inhibitor among the isolated alkaloids against both AChE and butyrylcholinesterase (BChE). Molecular docking revealed that rescinnamine interacted differently on AChE and BChE, by means of hydrophobic interactions and hydrogen bonding. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=Rauvolfia%20reflexa" title="Rauvolfia reflexa">Rauvolfia reflexa</a>, <a href="https://publications.waset.org/abstracts/search?q=indole%20alkaloids" title=" indole alkaloids"> indole alkaloids</a>, <a href="https://publications.waset.org/abstracts/search?q=acetylcholinesterase" title=" acetylcholinesterase"> acetylcholinesterase</a>, <a href="https://publications.waset.org/abstracts/search?q=butyrylcholinesterase" title=" butyrylcholinesterase"> butyrylcholinesterase</a>, <a href="https://publications.waset.org/abstracts/search?q=molecular%20docking" title=" molecular docking"> molecular docking</a> </p> <a href="https://publications.waset.org/abstracts/30034/cholinesterase-inhibitory-indole-alkaloids-from-the-bark-of-rauvolfia-reflexa" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/30034.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">593</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">11</span> Antioxidant Activity and Chemical Constituents of Leaf Essential Oils of Pseuduvaria Monticola and Pseuduvaria Macrophylla (Annonaceae)</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Hairin%20Taha">Hairin Taha</a>, <a href="https://publications.waset.org/abstracts/search?q=P.%20Narrima"> P. Pseuduvaria monticola is a rare montane forest species from the Annonaceae family. Traditionally, the plant was used to cure symptoms of fever, inflammation, stomach-ache and also to reduce the elevated levels of blood glucose. Scientifically, we have evaluated the antidiabetic potential of the Pseuduvaria monticola bark methanolic extract on certain in vitro cell based assays, followed by in vivo study. Results from in vitro models displayed PMm upregulated glucose uptake and insulin secretion in mouse pancreatic β-cells. In vivo study demonstrated the PMm down-regulated hyperglycaemia, oxidative stress and elevated levels of pro-inflammatory cytokines in type 2 diabetic rat models. Altogether, the study revealed that Pseuduvaria monticola might be used as a potential candidate for the management of type 2 diabetes and its related complications. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=type%202%20diabetes" title="type 2 diabetes">type 2 diabetes</a>, <a href="https://publications.waset.org/abstracts/search?q=Pseuduvaria%20monticola" title=" Pseuduvaria monticola"> Pseuduvaria monticola</a>, <a href="https://publications.waset.org/abstracts/search?q=insulin%20secretion" title=" insulin secretion"> insulin secretion</a>, <a href="https://publications.waset.org/abstracts/search?q=glucose%20uptake" title=" glucose uptake"> glucose uptake</a> </p> <a href="https://publications.waset.org/abstracts/13005/antidiabetic-potential-of-pseuduvaria-monticola-bark-extract-on-the-pancreatic-cells-nit-1-and-type-2-diabetic-rat-model" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/13005.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">439</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">9</span> New Quinazoline Derivative Induce Cytotoxic Effect against Mcf-7 Human Breast Cancer Cell</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Maryam%20Zahedi%20Fard">Maryam Zahedi Fard</a>, <a href="https://publications.waset.org/abstracts/search?q=Nazia%20Abdul%20Majid"> Nazia Abdul Majid</a>, <a href="https://publications.waset.org/abstracts/search?q=Hapipah%20Mohd%20Ali"> Hapipah Mohd Ali</a>, <a href="https://publications.waset.org/abstracts/search?q=Mahmood%20Ameen%20Abdulla"> Mahmood Ameen Abdulla</a> </p> <p class="card-text"><strong>Abstract:</strong></p> New quinazoline schiff base 3-(5-bromo-2-hydroxy-3-methoxybenzylideneamino)-2-(5-bromo-2-hydroxy-3-methoxyphenyl)-2,3-dihydroquinazolin-4(1H)-one was investigated for anticancer activity against MCF-7 human breast cancer cell line with involved mechanism of apoptosis. The compound demonstrated a remarkable antiproliferative effect, with an IC50 value of 3.41 ± 0.34, after 72 hours of treatment. Morphological apoptotic features in treated MCF-7 cells were observed by AO/PI staining. Furthermore, treated MCF-7 cells subjected to apoptosis death, as exhibited by perturbation of mitochondrial membrane potential and cytochrome c release as well as increase in ROS generation. We also found activation of caspases 3/7 and -9. Moreover, acute toxicity test demonstrated the nontoxic nature of the compound in mice. The structural elucidation of the isolated compounds has been performed using spectral methods such as UV, IR, MS, 1D, and 2D NMR. Rauvolfine B showed anti proliferation activity on HCT-116 cancer cell line, its cytotoxicity induction was observed using MTT assay in eight different cell lines. Annexin-V is serving as a marker for apoptotic cells and the Annexin-V-FITC assay was carried out to observe the detection of cell-surface Phosphatidylserine (PS). Apoptosis was confirmed by using caspase-8 and -9 assays. Cell cycle arrest was also investigated using flowcytometric analysis. rauvolfine B had exhibited significantly higher cytotoxicity against HCT-116 cell line. The treatment significantly arrested HCT-116 cells in the S phase. Together, the results presented in this study demonstrated that rauvolfine B inhibited the proliferation of HCT-116 cells and programmed cell death followed by cell cycle arrest. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=apocynacea" title="apocynacea">apocynacea</a>, <a href="https://publications.waset.org/abstracts/search?q=indole%20alkaloid" title=" indole alkaloid"> indole alkaloid</a>, <a href="https://publications.waset.org/abstracts/search?q=apoptosis" title=" apoptosis"> apoptosis</a>, <a href="https://publications.waset.org/abstracts/search?q=cell%20cycle%20arrest" title=" cell cycle arrest"> cell cycle arrest</a> </p> <a href="https://publications.waset.org/abstracts/13403/rauvolfine-b-isolated-from-the-bark-of-rauvolfia-reflexa-apocynaceae-induces-apoptosis-through-activation-of-caspase-9-coupled-with-s-phase-cell-cycle-arrest" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/13403.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">334</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">7</span> A Prenylflavanoid, HME5 with Antiproliferative Activity in Human Ovarian Cancer Cells</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Mashitoh%20Abd%20Rahman">Mashitoh Abd Rahman</a>, <a href="https://publications.waset.org/abstracts/search?q=Najihah%20Mohd%20Hashim"> Najihah Mohd Hashim</a>, <a href="https://publications.waset.org/abstracts/search?q=Faiqah%20Ramli"> Faiqah Ramli</a>, <a href="https://publications.waset.org/abstracts/search?q=Syam%20Mohan"> Syam Mohan</a>, <a href="https://publications.waset.org/abstracts/search?q=Noraziah%20Nordin"> Noraziah Nordin</a>, <a href="https://publications.waset.org/abstracts/search?q=Hamed%20Karimian"> Hamed Karimian</a>, <a href="https://publications.waset.org/abstracts/search?q=Hapipah%20Mohd%20Ali"> Hapipah Mohd Ali</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Ovarian cancer is the most lethal gynecological malignancies. HME5, a prenylflavanoid has been isolated from local medicinal plant. This compound has been reported to possess a broad spectrum of biological activities including anticancer property. However, the potential of HME5 as an antiproliferative and cytotoxic agent on an ovarian cancer cells has not yet been investigated. In this present study, we examined the antiproliferative and cytotoxic effect of HME5 on Caov-3 (Human Ovarian Adenocarcinoma) cell line by using 3-[4,5-dimethylthizol-2-y]-2,5-diphenyltetrazolium bromide (MTT) assay, Acridine orange and propidium Iodide (AOPi) and cell cycle analysis study. HME5 has shown to inhibit Caov-3 in a time-dependent manner with the IC50 values of 5µg/ml, 2µg/ml and 1µg/ml after 24h, 48h and 72h treatment, respectively. Morphological study from AOPi analysis showed that HME5 induced apoptosis after 24 and 48h post-treatment. Nevertheless, HME5 exhibited cell cycle arrest at G1 phase as indicated in flow cytometry cell cycle profiling. In conclusion, HME5 inhibited proliferation of Caov-3 through induction of apoptosis and cell cycle arrest at G1 phase. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=apoptosis" title="apoptosis">apoptosis</a>, <a href="https://publications.waset.org/abstracts/search?q=prenylflavanoid" title=" prenylflavanoid"> prenylflavanoid</a>, <a href="https://publications.waset.org/abstracts/search?q=ovarian%20cancer" title=" ovarian cancer"> ovarian cancer</a>, <a href="https://publications.waset.org/abstracts/search?q=HME5" title=" HME5 "> HME5 </a> </p> <a href="https://publications.waset.org/abstracts/13503/a-prenylflavanoid-hme5-with-antiproliferative-activity-in-human-ovarian-cancer-cells" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/13503.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">461</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">6</span> New Quinazoline Derivative Exhibit Cytotoxic Effect agaisnt MCF-7 Human Breast Cancer Cell</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Maryam%20Zahedifard">Maryam Zahedifard</a>, <a href="https://publications.waset.org/abstracts/search?q=Fadhil%20Lafta%20Faraj"> Fadhil Lafta Faraj</a>, <a href="https://publications.waset.org/abstracts/search?q=Nazia%20Abdul%20Majid"> Nazia Abdul Majid</a>, <a href="https://publications.waset.org/abstracts/search?q=Hapipah%20Mohd%20Ali"> Hapipah Mohd Ali</a>, <a href="https://publications.waset.org/abstracts/search?q=Mahmood%20Ameen%20Abdulla"> Mahmood Ameen Abdulla</a> </p> <p class="card-text"><strong>Abstract:</strong></p> The new quinazoline Schiff bases have been synthesized through condensation reaction of 2-aminobenzhydrazide with 5-bromosalicylaldehyde and 3-methoxy-5-bromosalicylaldehyde. The compound was investigated for anticancer activity against MCF-7 human breast cancer cell line. It demonstrated a remarkable antiproliferative effect, with an IC50 value of 3.41±0.34, after 72 hours of treatment. Most apoptosis morphological features in treated MCF-7 cells were observed by AO/PI staining. The results of cell cycle analysis indicate that compounds did not induce S and M phase arrest in cell after 24 hours of treatment. Furthermore, MCF-7 cells treated with compound subjected to apoptosis death, as exhibited by perturbation of mitochondrial membrane potential and cytochrome C release as well as increase in ROS generation. We also found activation of caspases 3/7 and -9. Moreover, acute toxicity results demonstrated the nontoxic nature of the compounds in mice. Our results showed the selected compound significantly induce apoptosis in MCF-7 cells via intrinsic pathway, which might be considered as a potential candidate for further in vivo and clinical breast cancer studies. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=quinazoline%20Schiff%20base" title="quinazoline Schiff base">quinazoline Schiff base</a>, <a href="https://publications.waset.org/abstracts/search?q=apoptosis" title=" apoptosis"> apoptosis</a>, <a href="https://publications.waset.org/abstracts/search?q=MCF-7" title=" MCF-7"> MCF-7</a>, <a href="https://publications.waset.org/abstracts/search?q=caspase" title=" caspase"> caspase</a>, <a href="https://publications.waset.org/abstracts/search?q=cell%20cycle" title=" cell cycle"> cell cycle</a>, <a href="https://publications.waset.org/abstracts/search?q=acute%20toxicity" title=" acute toxicity"> acute toxicity</a> </p> <a href="https://publications.waset.org/abstracts/14365/new-quinazoline-derivative-exhibit-cytotoxic-effect-agaisnt-mcf-7-human-breast-cancer-cell" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/14365.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">442</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">5</span> Studies on Induction of Cytotoxicity Through Apoptosis In Ovarian Cancer Cell Line (CAOV-3) by Chloroform Extract of Artocarpus Kemando Miq</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Noor%20Shafifiyaz%20Mohd%20Yazid">Noor Shafifiyaz Mohd Yazid</a>, <a href="https://publications.waset.org/abstracts/search?q=Najihah%20Mohd%20Hashim"> Najihah Mohd Hashim</a>, <a href="https://publications.waset.org/abstracts/search?q=Hapipah%20Mohd%20Ali"> Hapipah Mohd Ali</a>, <a href="https://publications.waset.org/abstracts/search?q=Syam%20Mohan"> Syam Mohan</a>, <a href="https://publications.waset.org/abstracts/search?q=Rosea%20Go"> Rosea Go</a> </p> <p class="card-text"><strong>Abstract:</strong></p> Artocarpus kemando is a plant species from Moraceae family. This plant is used as household utensil by the local and the fruits are edible. The plants’ bark was used for the extraction process and yielded the chloroform crude extract which was used to screen for anticancer potential. The cytotoxic effect of the extract on CAOV-3 and WRL 68 cell lines were determined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide or MTT assays. Qualitative AO/PI assay was performed to confirm the apoptosis and necrosis process. Meanwhile, the measurement of cell loss, nuclear morphology, DNA content, cell membrane permeability, mitochondrial membrane potential changes and cytochrome c release from mitochondria were detected through cytotoxicity 3 assay. In MTT assay, A. kemando inhibited 50% growth of CAOV-3 cells at 27.9 ± 0:03, 20.1± 0:03, 18.21± 0:04 µg/mL after 24, 48 and 72 hour, respectively. The morphology changes can be seen on CAOV-3 with a production of cell membrane blebbing, cromatin condensation and apoptotic bodies. Evaluation of cytotoxicity 3 on CAOV-3 cells after treated with extract resulting loss of mitochondrial membrane potential and release of cytochrome c from mitochondria. The chemical structures of both newly synthesized compounds (1 and 2) were confirmed by FT-IR and X-ray crystallography studies. The cytotoxic effect of compounds was investigated against MCF-7 human breast cancer cells. MTT results showed that (1) and (2) decreased the viability of MCF-7 cells in a time-dependent manner, exhibiting an IC50 value of 3.23 ± 0.28 µg/mL and 3.41 ± 0.34 µg/mL, respectively, after a 72-hours treatment period. In contrast, they did not show significant anti-proliferative effect towards MCF-10A normal breast cells and WRL-68 normal liver cells. We found a perturbation in mitochondrial membrane potential and increased cytochrome c release from the mitochondria to the cytosol, suggesting an activation of apoptosis by compounds, which was confirmed by activation of the initiator caspase-9 and the executioner caspases-3/7. (1) was also able to trigger extrinsic pathway via activation of caspase-8 and inhibition of NF-κB translocation. The acute toxicity test showed no toxicity effect of the compounds in rats. Our results showed that the selected synthesized compounds are highly potent to induce apoptosis in MCF-7 cells via either intrinsic or extrinsic mitochondrial pathway. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=Quinazoline%20Schiff%20base" title="Quinazoline Schiff base">Quinazoline Schiff base</a>, <a href="https://publications.waset.org/abstracts/search?q=apoptosis" title=" apoptosis"> apoptosis</a>, <a href="https://publications.waset.org/abstracts/search?q=MCF-7%20human%20breast%20cancer%20cell%20line" title=" MCF-7 human breast cancer cell line"> MCF-7 human breast cancer cell line</a>, <a href="https://publications.waset.org/abstracts/search?q=caspase" title=" caspase"> caspase</a>, <a href="https://publications.waset.org/abstracts/search?q=NF-%CE%BAB%20translocation" title=" NF-κB translocation"> NF-κB translocation</a> </p> <a href="https://publications.waset.org/abstracts/13587/synthesis-and-cytotoxic-activity-of-new-quinazolinone-based-compounds-against-human-breast-cancer-cell-line-mcf-7" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/13587.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">492</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">3</span> Induction of Cytotoxicity and Apoptosis in Ovarian Cancer Cell Line (CAOV-3) by an Isoquinoline Alkaloid Isolated from Enicosanthellum pulchrum (King) Heusden</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Noraziah%20Nordin">Noraziah Nordin</a>, <a href="https://publications.waset.org/abstracts/search?q=Najihah%20Mohd%20Hashim"> Najihah Mohd Hashim</a>, <a href="https://publications.waset.org/abstracts/search?q=Nazia%20Abdul%20Majid"> Nazia Abdul Majid</a>, <a href="https://publications.waset.org/abstracts/search?q=Mashitoh%20Abdul%20Rahman"> Mashitoh Abdul Rahman</a>, <a href="https://publications.waset.org/abstracts/search?q=Hamed%20Karimian"> Hamed Karimian</a>, <a href="https://publications.waset.org/abstracts/search?q=Hapipah%20Mohd%20Ali"> Hapipah Mohd Ali </a> </p> <p class="card-text"><strong>Abstract:</strong></p> Enicosanthellum pulchrum belongs to family Annonaceae is also known as family of 'mempisang' in Malaysia. Recent studies have been successful in ROS targeted therapies via antioxidants using mouse models in cancer therapeutics. The present study was designed to scrutinize anticancer activity, antioxidant activity of 5 different extracts of Thymus serpyllum in MDA-MB-231, MCF-7, HepG2, HCT-116, PC3, and A549. Identification of the phytochemicals present in the most active extract of Thymus serpyllum was conducted using gas chromatography coupled with mass spectrophotometry and antioxidant activity was measured by using DPPH radical scavenging and FRAP assay. Anticancer impact of the extract in terms of IC50 was evaluated using MTT cell viability assay. Results revealed that the hexane extract showed the best anticancer activity in HepG2 (Liver Carcinoma Cell Line) with an IC50 value of 23 ± 0.14 µg/ml followed by 25 µg/ml in HCT-116 (Colon Cancer Cell Line), 30 µm/ml in MCF-7 (Breast Cancer Cell Line), 35 µg/ml in MDA-MB-231 (Breast Cancer Cell Line), 57 µg/ml in PC3 (Prostate Cancer Cell Line) and 60 µg/ml in A549 (Lung Carcinoma Cell Line). GC-MS profile of the hexane extract showed the presence of 31 compounds with carvacrol, thymol and thymoquione being the major compounds. Phenolics such as Vitamin E, terpinen-4-ol, borneol and phytol were also identified. Hence, here we present the first report on cytotoxicity of hexane extract of Thymus serpyllum extract in HepG2 cell line with a robust anticancer activity with an IC50 of 23 ± 0.14 µg/ml. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=Thymus%20serpyllum%20L." title="Thymus serpyllum L.">Thymus serpyllum L.</a>, <a href="https://publications.waset.org/abstracts/search?q=hexane%20extract" title=" hexane extract"> hexane extract</a>, <a href="https://publications.waset.org/abstracts/search?q=GC-MS%20profile" title=" GC-MS profile"> GC-MS profile</a>, <a href="https://publications.waset.org/abstracts/search?q=antioxidant%20activity" title=" antioxidant activity"> antioxidant activity</a>, <a href="https://publications.waset.org/abstracts/search?q=anticancer%20activity" title=" anticancer activity"> anticancer activity</a>, <a href="https://publications.waset.org/abstracts/search?q=HepG2%20cell%20line" title=" HepG2 cell line"> HepG2 cell line</a> </p> <a href="https://publications.waset.org/abstracts/13474/hexane-extract-of-thymus-serpyllum-l-gc-ms-profile-antioxidant-potential-and-anticancer-impact-on-hepg2-liver-carcinoma-cell-line" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/13474.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">517</span> </span> </div> </div> <div class="card paper-listing mb-3 mt-3"> <h5 class="card-header" style="font-size:.9rem"><span class="badge badge-info">1</span> In vivo Antidiabetic and Antioxidant Potential of Pseudovaria macrophylla Extract</h5> <div class="card-body"> <p class="card-text"><strong>Authors:</strong> <a href="https://publications.waset.org/abstracts/search?q=Aditya%20Arya">Aditya Arya</a>, <a href="https://publications.waset.org/abstracts/search?q=Hairin%20Taha"> Hairin Taha</a>, <a href="https://publications.waset.org/abstracts/search?q=Ataul%20Karim%20Khan"> Ataul Karim Khan</a>, <a href="https://publications.waset.org/abstracts/search?q=Nayiar%20Shahid"> Nayiar Shahid</a>, <a href="https://publications.waset.org/abstracts/search?q=Hapipah%20Mohd%20Ali"> Hapipah Mohd Ali</a>, <a href="https://publications.waset.org/abstracts/search?q=Mustafa%20Ali%20Mohd"> Mustafa Ali Mohd</a> </p> <p class="card-text"><strong>Abstract:</strong></p> This study has investigated the antidiabetic and antioxidant potential of Pseudovaria macrophylla bark extract on streptozotocin–nicotinamide induced type 2 diabetic rats. LCMS-QTOF and NMR experiments were done to determine the chemical composition in the methanolic bark extract. For in vivo experiments, the STZ (60 mg/kg/b.w, 15 min after 120 mg/kg/1 nicotinamide, i.p.) induced diabetic rats were treated with methanolic extract of Pseuduvaria macrophylla (200 and 400 mg/kg∙bw) and glibenclamide (2.5 mg/kg) as positive control respectively. Biochemical parameters were assayed in the blood samples of all groups of rats. The pro-inflammatory cytokines, antioxidant status and plasma transforming growth factor βeta-1 (TGF-β1) were evaluated. The histological study of the pancreas was examined and its expression level of insulin was observed by immunohistochemistry. In addition, the expression of glucose transporters (GLUT 1, 2 and 4) were assessed in pancreas tissue by western blot analysis. The outcomes of the study displayed that the bark methanol extract of Pseuduvaria macrophylla has potentially normalized the elevated blood glucose levels and improved serum insulin and C-peptide levels with significant increase in the antioxidant enzyme, reduced glutathione (GSH) and decrease in the level of lipid peroxidation (LPO). Additionally, the extract has markedly decreased the levels of serum pro-inflammatory cytokines and transforming growth factor beta-1 (TGF-β1). Histopathology analysis demonstrated that Pseuduvaria macrophylla has the potential to protect the pancreas of diabetic rats against peroxidation damage by downregulating oxidative stress and elevated hyperglycaemia. Furthermore, the expression of insulin protein, GLUT-1, GLUT-2 and GLUT-4 in pancreatic cells was enhanced. The findings of this study support the anti-diabetic claims of Pseudovaria macrophylla bark. <p class="card-text"><strong>Keywords:</strong> <a href="https://publications.waset.org/abstracts/search?q=diabetes%20mellitus" title="diabetes mellitus">diabetes mellitus</a>, <a href="https://publications.waset.org/abstracts/search?q=Pseuduvaria%20macrophylla" title=" Pseuduvaria macrophylla"> Pseuduvaria macrophylla</a>, <a href="https://publications.waset.org/abstracts/search?q=alkaloids" title=" alkaloids"> alkaloids</a>, <a href="https://publications.waset.org/abstracts/search?q=caffeic%20acid" title=" caffeic acid"> caffeic acid</a> </p> <a href="https://publications.waset.org/abstracts/12490/in-vivo-antidiabetic-and-antioxidant-potential-of-pseudovaria-macrophylla-extract" class="btn btn-primary btn-sm">Procedia</a> <a href="https://publications.waset.org/abstracts/12490.pdf" target="_blank" class="btn btn-primary btn-sm">PDF</a> <span class="bg-info text-light px-1 py-1 float-right rounded"> Downloads <span class="badge badge-light">357</span> </span> </div> </div> </div> </main> <footer> <div id="infolinks" class="pt-3 pb-2"> <div class="container"> <div style="background-color:#f5f5f5;" class="p-3"> <div class="row"> <div class="col-md-2"> <ul class="list-unstyled"> About <li><a href="https://waset.org/page/support">About Us</a></li> <li><a href="https://waset.org/page/support#legal-information">Legal</a></li> <li><a target="_blank" rel="nofollow" href="https://publications.waset.org/static/files/WASET-16th-foundational-anniversary.pdf">WASET celebrates its 16th foundational anniversary</a></li> </ul> </div> <div class="col-md-2"> <ul class="list-unstyled"> Account <li><a href="https://waset.org/profile">My Account</a></li> </ul> </div> <div class="col-md-2"> <ul class="list-unstyled"> Explore <li><a href="https://waset.org/disciplines">Disciplines</a></li> <li><a href="https://waset.org/conferences">Conferences</a></li> <li><a href="https://waset.org/conference-programs">Conference Program</a></li> <li><a href="https://waset.org/committees">Committees</a></li> <li><a href="https://publications.waset.org">Publications</a></li> </ul> </div> <div class="col-md-2"> <ul class="list-unstyled"> Research <li><a href="https://publications.waset.org/abstracts">Abstracts</a></li> <li><a href="https://publications.waset.org">Periodicals</a></li> <li><a href="https://publications.waset.org/archive">Archive</a></li> </ul> </div> <div class="col-md-2"> <ul class="list-unstyled"> Open Science <li><a target="_blank" rel="nofollow" href="https://publications.waset.org/static/files/Open-Science-Philosophy.pdf">Open Science Philosophy</a></li> <li><a target="_blank" rel="nofollow" href="https://publications.waset.org/static/files/Open-Science-Award.pdf">Open Science Award</a></li> <li><a target="_blank" rel="nofollow" href="https://publications.waset.org/static/files/Open-Society-Open-Science-and-Open-Innovation.pdf">Open Innovation</a></li> <li><a target="_blank" rel="nofollow" href="https://publications.waset.org/static/files/Postdoctoral-Fellowship-Award.pdf">Postdoctoral Fellowship Award</a></li> <li><a target="_blank" rel="nofollow" href="https://publications.waset.org/static/files/Scholarly-Research-Review.pdf">Scholarly Research Review</a></li> </ul> </div> <div class="col-md-2"> <ul class="list-unstyled"> Support <li><a href="https://waset.org/page/support">Support</a></li> <li><a href="https://waset.org/profile/messages/create">Contact Us</a></li> <li><a href="https://waset.org/profile/messages/create">Report Abuse</a></li> </ul> </div> </div> </div> </div> </div> <div class="container text-center"> <hr style="margin-top:0;margin-bottom:.3rem;"> <a href="https://creativecommons.org/licenses/by/4.0/" target="_blank" class="text-muted small">Creative Commons Attribution 4.0 International License</a> <div id="copy" class="mt-2">© 2024 World Academy of Science, Engineering and Technology</div> </div> </footer> <a href="javascript:" id="return-to-top"><i class="fas fa-arrow-up"></i></a> <div class="modal" id="modal-template"> <div class="modal-dialog"> <div class="modal-content"> <div class="row m-0 mt-1"> <div class="col-md-12"> <button type="button" class="close" data-dismiss="modal" aria-label="Close"><span aria-hidden="true">×</span></button> </div> </div> <div class="modal-body"></div> </div> </div> </div> <script src="https://cdn.waset.org/static/plugins/jquery-3.3.1.min.js"></script> <script src="https://cdn.waset.org/static/plugins/bootstrap-4.2.1/js/bootstrap.bundle.min.js"></script> <script src="https://cdn.waset.org/static/js/site.js?v=150220211556"></script> <script> jQuery(document).ready(function() { /*jQuery.get("https://publications.waset.org/xhr/user-menu", function (response) { jQuery('#mainNavMenu').append(response); });*/ jQuery.get({ url: "https://publications.waset.org/xhr/user-menu", cache: false }).then(function(response){ jQuery('#mainNavMenu').append(response); }); }); </script> </body> </html>
